摘    要:目的 探讨E-cadherin基因在人非小细胞肺癌组织(NSCLC)中的表达及其意义.方法 采用实时荧光定量PCR(Q-PCR)方法检测41例NSCLC组织、9例癌旁组织、9例良性病变肺组织中E-cadherin mRNA表达的相对水平,分析其表达与临床、组织病理学特征的关系.结果 E-cadherin mRNA在NSCLC组织表达水平为(9.97±4.25),低于癌旁组织的(11.77±1.91)及良性疾病组织的(23.11±3.52),差异有统计学意义(P<0.01).在NSCLC组织中E-cadherin mRNA的表达与淋巴结转移有关(P<0.01),与患者年龄、性别、吸烟史、病理类型、分化程度及临床分期无关(P>0.05).结论 E-cadherin mRNA表达下调与非小细胞肺癌的浸润和转移可能呈负相关,是评价NSCLC进展的一个潜在指标.

Abstract:Objective To explore the expression of E-cadherin genes and its significance in human non-small cell lung cancer(NSCLC)tissues.Methods The real-time fluorescence quantitative polymerase chain reaction(Q-PCR) was used to detect the relative expression levels of E-cadherin mRNA in 41 cases of NSCLC tissues,nine cases of paracarcinoma tissues and nine cases of lung benign lesions,the relationship between their expressions and clinical and pathologic characters was analyzed.Results The average expression level of E-cadherin mRNA in NSCLC tissues was lower than that of paracarcinoma tissues(9.97±4.25 vs 11.77±1.91,P<0.01) and that of lung benign lesions(23.11±3.52),which had statistical difference(P<0.01).The expression of E-cadherin mRNA was correlated closely with lymph node metastasis in NSCLC tissues(P<0.01),but was not correlated with patients′ age,sex,smoking history,pathological types,grade of differentiation and clinical stages(P>0.05).Conclusion The decrease of expression of E-cadherin may has a negative correlation with invasion and metastasis of NSCLC,which may be one of potential indexes in the evaluation of NSCLC progress.
