Toll样受体TRIF信号因子在乳腺癌中的表达与临床关系

摘    要:目的观察Toll样受体信号TRIF(TIR结构域接头分子,TIR-domain-containing adapter-inducing interferon-β,TRIF)在不同分期乳腺癌中及癌旁组织中的表达,为进一步深入研究先天免疫下游信号因子与乳腺癌发病机制中的关系提出初步依据。方法收集35例乳腺癌患者的一般情况和临床病理特征等资料,应用RT-PCR、Western blot方法检测TRIF在乳腺癌及癌旁组织中的基因和蛋白表达水平。同时应用免疫组化检测TRIF在乳腺癌及癌旁组织结构中的分布,以观察在该组织中的表达特点。结果 RT-PCR和Western blot结果显示,与癌旁组织中相比,乳腺癌组织中TRIF基因和蛋白表达水平具有显著差异(P<0.01);Ⅰ期、Ⅱ期及Ⅲ期相比,TRIF的表达水平差异也具有显著性,且分期越晚表达越强(P<0.01);癌细胞及炎性细胞、浆细胞、纤维母细胞等在细胞核和细胞浆中均有强烈高表达和分布,不同细胞类型差异性表达TRIF,且与肿瘤的TNM分期、肿瘤大小、淋巴侵袭转移密切相关。结论 TRIF的表达与乳腺肿瘤的增殖和分化密切相关。

Abstract:The study was designed to evaluate the expression of TIR-domain-containing adapter-inducing interferon-β(TRIF) in cancer and adjacent tissues of breast carcinoma at different clinical stages,for benefited further study of the relationship between innate immune downstream signaling factors and breast carcinoma pathogenesis.Total of 35 breast cancer tissues were collected.HE staining,Immunohistochemistry,PCR and Western blot were used to measure the expression of TRIF in cancer and adjacent tissues of different clinical stages.TRIF expression in breast cancer tissue was significantly higher than that in paracancerous tissue(P < 0.01),and TRIF were differently expressed in clinical stage I,II and III,the later stage the stronger expression(P < 0.01).Furthermore,the expressions of TRIF were different in cells of carcinoma and paracancerous tissues and were closely related to TNM stages,the tumor size,and lymphnode metastasis.The expression of TRIF in carcinoma and immunocytes reveals the participation of innate immunity in breast carcinomas.
