反义p21WAF1/CIP1基因转染对肾癌细胞凋亡抑制作用的影响

摘    要:目的 探讨p21^WAF1/CIP1基因在顺铂诱导的GRC-1肾癌细胞凋亡过程中的作用。方法 使用脂质体转染方法将正义p21^WAF1/CIP1基因及反义p21^WAF1/CIP1基因基因分别导入GRC-1肾癌细胞系中,对所获转染细胞进行形态学观察和流式细胞仪检测。结果 在正义p21^WAF1/CIP1基因基因转染后GRC-1肾癌细胞中,顺铂诱导细胞凋亡的作用增强;在反义p21^WAF1/CIP1基因基因转染后GRC-1肾癌细胞中,顺铂诱导细胞凋亡的作用受到抑制。结论 在GRC-1肾癌细胞中,顺铂诱导的细胞凋亡至少部分是通过p21^WAF1/CIP1基因基因发挥作用。p21^WAF1/CIP1基因基因作为一个与细胞凋亡的相关基因可能参与肾癌细胞凋亡的调控。

Abstract:AIM To explore effect of p21 WAF1/CIP1 gene on GRC-1 renal carcinoma cell apoptosis mediated by Cisplatin. METHODS p21 WAF1/CIP1 and antisense p21 WAF1/CIP1 were respectively transfected into GRC-1 renal carcinoma cell line byusing lipofectin method. The observation of morphology and the detection of flow cytometer were made on transfected GRC-1 cells. RESULTS Apoptosis mediated by Cisplatin were reinforced on p21 WAF1/CIP1 gene transfected GRC-1 cells,but were inhibited on antisense p21 WAF1/CIP1 gene transfected GRC-1 cells. CONCLUSION p21 WAF1/CIP1 gene at least partly produces a marked effect on apoptosis mediated by Cisplatin. p21 WAF1/CIP1, as a gene closely related to apoptosis, might modulate the death or survivorship of renal carcinoma cell.
