改良线栓法制备大鼠大脑中动脉阻塞模型

摘    要:目的总结改良的、简单实用的大鼠大脑中动脉阻塞(MCAO)模型建立方法的相关经验。方法16只成年健康雄性Sprague Dawley(SD)大鼠,随机分为实验组(8只)和对照组(8只)。纵行切开左侧颈部旁正中皮肤,采用从左侧颈总动脉(CCA)插入尼龙栓线的制模方法,实验组术中分别使用显微动脉夹暂时夹闭CCA及颈内动脉。对照组仅暴露左侧大脑中动脉而不予其他处理。两组实验动物分别在缺血后24h进行神经功能评分、MRI常规轴面T_2W_1扫描、病理学检查以及四氮唑(TTC)染色判定模型成功与否。结果对照组MRI常规轴面T_2W_1扫描未发现异常,实验组MCAO后24h MRI T_2WI左侧基底节和顶叶皮层均显示异常高信号;对照组大鼠脑组织神经细胞结构正常,实验组大鼠脑组织神经细胞缺血24h时出现典型的缺血性神经细胞改变;两组实验动物神经功能评分差异有统计学意义(P〈0.05);实验组TTC染色结果均显示左侧额颞顶皮层及基底节区缺血。结论根据改良线拴法的相关经验制备大鼠MCAO模型,制模成功率高、重复性好、手术时间短,可作为一种较为理想的制作大鼠脑缺血模型的方法。

Experience of establishing rats brain artery occlusion of the model in improved method

Abstract:Objective To establish a simple and practical method of rat ischemia model. Methods 16 Male Sprague Dawley ( SD ) rats were randomly divided into experimental group (8)and control group (8), longitudinal median skin incision on the left side of the neck, using the common carotid artery(CCA) into the molding of nylon filament method, the experimental group were intraoperative temporary folder using micro artery clipping left CCA. The control group exposed to the left middle cerebral artery and no other treatment. Two groups of animals were carried out at 24 h after ischemic neurological score, MRI T_2W_1 scans, pathological examination and Tetrazolium (TTC) staining to determine the success of the model. Results The control group, MRI T_2_W1 scans were normal, in the experimental group after 24 h MRI T_2W_1 MCAO left basal ganglia and parietal cortex show abnormal high signal; The control group, normal structure of nerve cells in brain tissue, brain tissue of experimental rats 24 h nerve ceils lacking the typical ischemic nerve cell changes; Two groups of nerve function score was significantly different (P 〈0.05 ); Experi- mental group TTC staining results were top of the left frontal and temporal cortex and basal ganglia ischemia. Conclusions Pre- pared under the modified line tied to experience in rat MCAO model high success rate, reproducibility, shorter operative time, can be produced as an ideal method of cerebral ischemia.
