胚鼠大脑皮质一氧化氮合酶阳性神经元的体外生长发育研究

摘    要:本文在体外培养条件下观察了孕 14 d SD胚鼠大脑皮质中一氧化氮合酶阳性神经元的形态和发育过程。将孕 14 d SD胚鼠大脑皮质细胞悬液接种于 2 4孔培养板 ,实验按培养龄分 4组 ,分别在接种后 5d、10 d、15d和 2 0 d终止培养 ,继而进行NA DPH - d组织化学染色 ,观察各组一氧化氮合酶阳性神经元的数量和形态。结果显示 :培养后 5d即有一定数量的一氧化氮合酶阳性神经元出现 ,在 4个实验组中 ,以 15d组的一氧化氮合酶阳性神经元的数量为最多、突起最长 ,深染细胞数量也最多 ,大部分一氧化氮合酶阳性神经元的胞体与突起均局限分布在细胞团块内 ,胞体大小不一 ,只有少量的一氧化氮合酶阳性细胞散在分布在细胞团块之间 ,其突起伸向或围绕团块。本研究结果表明 ,在胚鼠大脑皮质的神经元内即已有一氧化氮合酶存在 ,提示一氧化氮可能与胚脑的生长发育有关

Abstract:The morphology and development of NOS positive neurons in the cerebral cortex of E14 rat were observed in vitro. Cerebral cortex cell suspension obtained from E14 rat were plated on 24 well culture plates. The experiments were divided into four groups according to the culture day. Cultures were terminated respectively at 5 d, 10 d, 15 d and 20 d after plating, then were stained by NADPH d histochemical method, and observed the number and shape about the NOS positive neurons of each group. The results were as follows: A certain number of NOS positive neurons were present already at the fifth day of culture, among those four groups, the numbers of the NOS positive neurons in the 15 d old culture was the most of all, their neurites were the longest, and the number of the intensely stained cells was also much more than in other groups. A major proportion of the NOS positive neuron cell bodies and neurites were distributed in the cell masses. The cell bodies were different in size, only a small proportion of the cells was scattered between the cell masses, but their neurites orientating or surrounding the cell mass. These results indicated that NOS positive neurons were present in the embryonic rat cerebral cortex suggesting the possibility that NO has relations to the development of embryonic brain.
