The Regulation of Secretion of Mesenchymal Stem Cells on Cytokines in Patients with Rheumatoid Arthritis

Objective To investigate the regulatory effect of mesenchymal stem cell secretions on cytokines in patients with rheumatoid arthritis(RA),in order to identify an effective way to alleviate the synovial inflammation of rheumatoid arthritis and reduce the production of inflammatory cytokines.Methods 49 patients with rheumatoid arthritis were randomly selected as the research objects,and epidemiological information of these patients was collected.The antibody levels of TNF-α,IL-1,IL-6 and IL-8 secreted by peripheral blood lymphocytes of patients stimulated by mesenchymal stem cell secretion were detected by ELISA,and the gene expression level of TNF-α,IL-1,IL-6,and IL-8 in peripheral blood of patients stimulated by mesenchymal stem cell secretion was detected by qPCR.Results The secretion of mesenchymal stem cells could significantly reduce the serum TNF-αantibody level of patients with rheumatoid arthritis,and the antibody levels of IL-1,IL-6 and IL-8 were also decreased;The gene expression levels of TNF-α,IL-1,IL-6 and IL-8 in the peripheral blood mononuclear cells of patients with rheumatoid arthritis were significantly decreased after co-culture of mesenchymal stem cells secretion and peripheral blood mononuclear cells of patients with rheumatoid arthritis for 24 hours(P<0.05).Conclusion The secretion of mesenchymal stem cells can regulate cytokines in patients with rheumatoid arthritis.
