Study on protective effect of eleven formula medicinal liquid on rats with acute soft tissue injury

Objective:To investigate the protective effect and mechanism of eleven formula medicinal liquid on rats with acute soft tissue injury caused by heavy objects.Methods:A model of acute soft tissue injury induced by heavy object in rats was established and randomly divided into model group,positive group(Voltaren 500 mg/kg)and high,medium and low dose groups of eleven formula medicinal liquid with 965 mg/kg and 483 mg,241mg/kg,respectively,with 8 rats in each group.Another 8 rats were selected as blank group.The next day after the modeling,the rats in each group were smeared and administered in vitro,twice a day,continuously administered for 3 days.Before and after administration,the diameter of the thigh muscles of the left and right hind limbs of each rat was measured with a vernier caliper,and the swelling degree was calculated.The soft tissue at the injury site was taken to prepare 10%tissue homogenate,and IL-1β,IL-6,TNF-α,PGE₂ factor levels were determined by ELISA separately,MDA content was detected by TBA method,SOD activity was detected by WST-1 method.Another part of soft tissue was taken,fixed with 4%paraformaldehyde,and HE slices were made to observe the pathological changes of soft tissue.Results:Eleven formula medicinal liquid could relieve the swelling of injured rats,inhibit the levels of IL-1β,IL6,TNF-α,PGE₂,MDA,and increase the activity of SOD,with significant differences compared with the model group(P<0.05 or P<0.01).It can improve the degree of muscle bundle disorder,muscle fibrinolysis,inflammatory cell infiltration and necrosis caused by injury.Conclusion:It has obvious protective effect on acute soft tissue injury in rats with eleven formula medicinal liquid.
