宫颈癌中COX-2与p53、E-cadherin蛋白表达的关系

摘    要:目的 探讨宫颈癌组织中环氧合酶 2 (COX 2 )的表达与 p5 3、E cadherin蛋白表达的关系。 方法 采用免疫组织化学S P法检测 4 1例宫颈癌和 10例正常宫颈上皮组织中COX 2、p5 3、E cadherin蛋白的表达水平。结果 宫颈癌组织中COX 2、p5 3表达水平明显高于正常宫颈上皮 (P <0 .0 1) ,而E cadherin蛋白的表达水平明显低于正常宫颈上皮 (P <0 .0 1) ;COX 2的高表达与宫颈癌淋巴结转移和浸润深度有关 (P <0 .0 5 ) ,与患者年龄、临床分期、组织学类型、分化程度无关 (P >0 0 5 ) ;COX 2表达阳性组 p5 3的表达水平明显高于COX 2表达阴性组 (P <0 0 5 ) ,COX 2表达阳性组的E cadherin蛋白的表达水平则低于相对应阴性组 ,差异有显著性 (P <0 0 5 )。结论 在宫颈癌的发生发展中COX 2、p5 3、E cadherin可能起重要作用。COX 2通过使抑癌基因p5 3失活 ,降低细胞黏附分子E cadherin的水平促进宫颈癌的发生。

Abstract:Purpose To investigate the expression of COX-2 and its relationship with p53 and E-cadherin expression. Methods The expression of COX-2 ,p53, E-cadherin protein in 41 cases of cervical carcinomas and 10 cases of normal cervical epithelia was examined by immunohistochemistry S-P method. Results The expression of COX-2 and p53 was significantly higher in cervical carcinomas than in normal cervical epithelia,the expression of E-cadherin was significantly lower in cervical carcinomas than in normal cervical epithelia(P<0.01).Overexpression of COX-2 in cervical carcinomas was related to lymph node metastasis and depth of invasion,but had no relation to age,FIGO staging,histopathological types and differentiation degree(P>0.05).The p53 was significantly higher in the COX-2-positive tumors than those in the negative ones, The E-cadherin was significantly lower in the COX-2-positive tumors than in the negative ones(P<0.05). Conclusions COX-2,p53,E-cadherin may be involved in carcinogenesis of cervical carcinoma. COX-2 may contribute to the tumorigenesis of cervical carcinoma by inactivation of p53 gene and down-regulation of E-cadherin protein.
