维吾尔族和汉族女性乳腺癌与KIAA1199基因表达调控的关系

目的探讨维吾尔族和汉族女性乳腺癌与KIAA1199表达调控的关系。方法收集维吾尔族和汉族女性乳腺纤维腺瘤、乳腺癌患者的石蜡包埋组织标本150例(维吾尔族68例,汉族82例),另选取新鲜组织标本58例(维吾尔族21例,汉族37例),采用免疫组织化学和逆转录聚合酶链反应(RT-PCR)法检测KIAA1199蛋白和mRNA表达水平。结果乳腺纤维腺瘤细胞中KIAA1199蛋白表达上调率为31.9%,乳腺癌细胞中KIAA1199蛋白表达上调率为76.7%,差异有统计学意义(P<0.01),维吾尔族、汉族女性乳腺癌KIAA1199表达上调率的变化趋势具有共性(维吾尔族2=12.30,P<0.01;汉族2=15.19,P<0.01),其族群差异无统计学意义(P>0.05)。KIAA1199基因的转录表达水平在乳腺癌组织中明显上调,其mRNA相对含量为0.967±0.281,乳腺纤维腺瘤组织中KIAA1199 mRNA含量为0.475±0.176,差异有统计学意义(P<0.01)。结论 KIAA1199表达上调伴随着乳腺病变进程,可能成为乳腺癌分子预警指标。

Association of breast cancer of Uighur and Han women with the KIAA1199 expression

Objective To investigate the association of the regulation of KIAA1199 gene expression at the mRNA and protein level with the breast cancer,and its relation to ethnic differences between Uighur and Han women.Methods This study collected 150 cases of paraffin-embedded breast lesion tissue specimens（Uighur 68 cases,Han 82 cases） and 58 cases of fresh breast lesion tissues from Uighur and Han women（Uighur 21 cases,Han 37 cases） with breast cancer or breast fibroadenoma,and analyzed the protein and mRNA expression of KIAA1199 by immunohistochemistry and semi-quantitative RT-PCR.Results（1） KIAA1199 protein was expressed at normal levels in breast fibroadenoma epithelial cells,and upregulated in a few cases（31.9%）,but the high rate of KIAA1199 protein expression was markedly increased in breast cancer（76.7%）,and the difference is significant among groups（P 〈 0.01）.The alteration trend of the high rate of KIAA1199 is common to women from both Uighur and Han ethnic groups,and the difference between two ethnic groups is also no statistically significant（P 〉 0.05）.（2） A significant increase in mRNA expression level（0.967 ± 0.281） was detected for KIAA1199 in breast cancer compared with breast fibroadenoma epithelial cells（0.475 ± 0.176）（P 〈 0.01）.Conclusion Upregulate of KIAA1199 expression may closely associated with breast cancer,and KIAA1199 may become a marker for the breast pathogenesis.
