针刺对局灶性脑缺血大鼠血糖和胰岛素水平的影响

摘    要:目的:观察针刺对局灶性脑缺血大鼠血糖和胰岛素水平的影响,探讨胰岛素的脑保护机制,为针刺防治缺血性脑血管疾病提供实验依据。方法:雄性Wistar大鼠随机分为正常组、假手术组、模型组、针刺组,每组40只,每组再分为6h、1d、3d、7d、14d5个亚组,每组8只。采用线栓法制作局灶性脑缺血模型。电针双侧"内关"穴和"曲池"穴,每天1次。采用Zea Longa神经体征评分评价大鼠神经功能缺损情况,激光多普勒检测大鼠软脑膜微循环血流量,血糖仪测量血糖,放射性免疫法检测大鼠血清胰岛素的含量。结果:针刺治疗3、7d后针刺组大鼠神经体征评分明显低于模型组(P<0.05)。针刺7、14d组大鼠软脑膜微循环血流量明显高于模型组和针刺1d组(P<0.05)。模型组及针刺各组大鼠造模后血糖水平和胰岛素水平显著低于正常组和假手术组(P<0.05)。针刺14d组大鼠血糖高于模型组(P<0.05)。从针刺1d后,各时间点针刺组大鼠胰岛素水平较模型组显著升高(P<0.05)。结论:针刺可以上调局灶性脑缺血大鼠血清胰岛素水平,发挥脑保护作用,且这种脑保护作用与血糖水平无明显相关性。

Effects of Electroacupuncture Intervention on Neurological Function,Blood Glucose and Insulin Levels in Rats with Focal Cerebral Ischemia

Abstract:Objective To observe the effect of electroacupuncture(EA)intervention on dynamic changes of neurological function,blood glucose and insulin levels in rats with focal cerebral ischemia(CI),so as to explore its mechanism underlying improvement of ischemic cerebral vascular disease.Methods Male Wistar rats were randomly divided into four groups:normal, sham-operation(sham),model and EA.CI model was induced by middle cerebral artery occlusion(MCAO).The four groups were further randomized into 5subgroups according to time-points of MCAO:6hour,day 1,day 3,day 7and day 14(8rats/ group).EA(2Hz/15Hz,1mA)was applied to bilateral"Neiguan"(PC 6)and"Quchi"(LI 11)for 30min,once daily for 1day to 14days.Neurological impairment was evaluated by Zea Longa 5-point scoring system.Local cerebral blood flow(LCBF)of the cerebral pia mater was determined using Laser Doppler Flowmetry.The levels of abdominal cavity venous blood glucose and serum insulin were measured by using aglucose meter and radioimmunoassay,respectively.Results In comparison with the normal and sham groups,the rats' body weights and LCBF levels at time-points of day 1,3,7and 14,and blood glucose and serum insulin levels at most time-points of 6h,day 1,3,7and 14after CI were significantly decreased in the model group(P<0.05), and neurological scores at time-points of 6h,day 1,3,7and 14were markedly increased in the model group(P<0.05).After EA intervention,compared with the model group,the rats' body weights at time-points of day 3,7and 14,LCBF levels on day 7 and 14,blood glucose on day 14,and serum insulin contents at time-points of day 1,3,7and 14were considerably increased in the EA group(P<0.05),while Zea Longa scores at time-points of day 3and 7were evidently decreased in the EA group(P<0.05).Conclusion EA intervention is effective in improving neurological function in CI rats,which is probably associated with its functions in improving cerebral blood flow,and up-regulating blood glucose and insulin levels.
