基于网络药理学研究黄连-黄柏药对治疗溃疡性结肠炎的作用机制

摘    要:目的:运用网络药理学技术研究黄连-黄柏药对治疗溃疡性结肠炎的主要活性成分,预测活性成分的作用靶点、建立“药对-活性成分-疾病-作用靶点”网络,探讨黄连-黄柏药对治疗溃疡性结肠炎的潜在作用机制。方法:运用中药系统药理学数据库与分析平台(TCMSP)筛选黄连-黄柏药对的化学成分,借助人类基因数据库获取溃疡性结肠炎的相关靶点基因,将药物作用靶点及疾病相关靶标基因取交集,使用STRING数据库对关键作用靶点构建蛋白质-蛋白质相互作用(PPI)网络,通过Cytoscape软件将结果可视化,借助DAVID数据库对“黄连-黄柏”治疗溃疡性结肠炎相关靶基因基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)富集分析。结果:经过筛选出黄连-黄柏药对主要关键靶点蛋白34个和分子信号通路30个,包含PTGS2、IL6、CXCL8等关键基因及T细胞受体、核因子κB、NOD样受体、Toll样受体信号通路。结论:黄连-黄柏药对治疗溃疡性结肠炎是多靶点、多个信号通路共同调节的复杂过程,预测其潜在作用机制为深入研究清热法治疗溃疡性结肠炎的作用机制提供理论依据。

Abstract:Objective:To apply network pharmacology research of couple medicine Rhizoma Coptidis and Cortex Phellodendri the main active ingredients in the treatment of ulcerative colitis,prediction of active ingredients targets,establish of“medicine-the active ingredient-disease-targets”,further discusses the couple medicine Rhizoma Coptidis and Cortex Phellodendri potential mechanisms for the treatment of ulcerative colitis.Methods:Traditional Chinese Medicine Systems Pharmacology and Analysis Platform was used to screen the chemical constituents of the couple medicine Rhizoma Coptidis and Cortex Phellodendri.The relevant target genes of ulcerative colitis(UC)were obtained with the help of human Gene database,and the key action targets of the couple medicine Rhizoma Coptidis and Cortex Phellodendri in the treatment of ulcerative colitis were obtained by taking the intersection of drug action targets and disease-related target genes.The STRING database was used to construct protein interaction networks for key targets,and the results were visualized by using Cytoscape software.The DAVIDdatabase was used to analyze the functional and metabolic pathways of target genes related to the treatment of ulcerative colitis in the couple medicine Rhizoma Coptidis and Cortex Phellodendri.Results:A total of 34 key target proteins and 30 molecular signaling pathways for the couple medicine Rhizoma Coptidis and Cortex Phellodendri were screened.Main key genes such as PTGS2,IL6,CXCL8,and T cell receptor signaling pathway,NF-κB signaling pathway,NOD-like receptor signaling pathway and Toll-like receptor signaling pathway.Conclusion:The treatment of ulcerative colitis with the couple medicine Rhizoma Coptidis and Cortex Phellodendri is a complex process of multiple targets and multiple signaling pathways.The prediction of the potential mechanism provides theoretical basis for further study of the mechanism of action of the heat-clearing method in the treatment of ulcerative colitis.
