荆花胃康胶丸对实验性胃溃疡大鼠粘膜愈合和EGF、EGFR表达的影响

目的通过荆花胃康胶丸对血清和胃粘膜组织的EGF含量及胃粘膜上皮细胞EGFR表达的影响,探讨该药对实验性胃溃疡大鼠胃粘膜的保护作用及其作用机制。方法采用冰醋酸注射至大鼠浆膜下形成胃溃疡模型,随机分成生理盐水(B)组,雷尼替丁(C)组,荆花胃康胶丸10 mg.kg-1.d-1(D)组、15 mg.kg-1.d-1(E)组和20 mg.kg-1.d-1(F)组,设立正常对照(A)组,用游标卡尺测量溃疡指数(UI),用放射免疫法检测血清及组织EGF含量,用免疫组化法检测胃粘膜组织EGFR的表达。结果C、D、E、F组与B组比较、E、F组与C组比较,UI、EGF含量均有显著性差异(P<0.05或0.01);D、E、F组间也有差异(P<0.05);C、D、E、F组溃疡边缘组织EGFR阳性细胞表达较A、B组逐渐增多(P<0.01)。D、E、F组间EGFR表达也有显著性差异(P<0.05)。结论荆花胃康胶丸通过促进EGF分泌、促进或稳定胃粘膜上皮细胞EGFR表达,加速上皮细胞增殖、分化、迁移,组织修复而加速溃疡愈合、提高溃疡愈合质量,改善溃疡预后。

Effect of Jinhua Weikang Soft Gelatin Capsule on concrescence of gastric mucosa and expression of EGF and EGFR in rats with gastric ulcer constructed with experiment

Objective To investigate protective effects and mechanism of Jinhua Weikang Soft Gelatin Capsule on concrescence of gastric mucosa and expression of EGF and EGFR in rats with gastric ulcer constructed with experiment.Methods The model of gastric ulcer was constructed by injection of acetic acid in subserosa.The rats were randomly divided into: 0.9% NaCl 10 mg·kg~(-1)·d~(-1) group(B),Ranitidine 30 mg·kg~(-1)·d~(-1) group(C),Jinhua Weikang soft gelatin capsule 10 mg·kg~(-1)·d~(-1) group(D),Jinhua Weikang soft gelatin capsule 15 mg·kg~(-1)·d~(-1) group(E),Jinhua Weikang soft gelatin capsule 20 mg·kg~(-1)·d~(-1) group(F).Normal control group(A).The index of ulcer(UI) was measured by vernier caliper,and the levels of EGF were examined by radioimmunity.The expression of EGFR in the mucosa around the ulcer was detected by SABC kit immunohistochemical method and MPIAS microimage analysis system.Results UI and EGF in group C、D、E and F were significant difference compared with group B,UI and EGF in group E and F were significant difference compared with group C(P<0.05 or <0.01).There were difference of UI and EGF among group D,E and F(P<0.05).Positive expression of EGFR around ulcerative tissues in group C,D,E and F were more than that in group A and B(P<0.01).There was significant difference of EGFR among group of D,E and F(P<0.05).Conclusion Jinhua Weikang soft gelatin capsule can improve healing of ulcer and prognosis of ulcer by promoting secretion of EGF,promoting or stabilizing expression if EGFR in epithelial cells,increasing proliferation,differentiation,migration and tissue repairing.
