水蛭提取物对血管内皮细胞VEGF受体-2表达的影响

摘    要:目的:研究水蛭提取物对凝血酶诱导的RF-6A细胞(恒河猴视网膜脉络膜血管内皮细胞)跨膜受体VEGFR2表达的影响,从而探讨中药水蛭用于防治新生血管形成中的作用。方法:将培养细胞分成水蛭组,凝血酶组,凝血酶+水蛭组及空白组。在前期实验基础上选择64g/L水蛭提取液及20NIHU/mL凝血酶。8h后,采用免疫组织化的方法检测各组细胞VEGFR2的表达,并进行图像分析。结果:免疫组化分析:与空白组比较水蛭组RF-6A细胞膜上VEGFR2的表达明显降低,降低程度高于凝血酶组,小于混合组,Sig=0.000,P<0.01各组间的差异有统计学意义。结论:水蛭提取物可使内皮细胞跨膜受体VEGFR2表达明显减少,推测其对VEGF受体介导的跨膜信号传导途径的作用可能是通过竞争性地抑制VEGF与其受体的结合。
