乙型肝炎病毒X基因部分区段及其编码氨基酸变异的分析

摘    要:目的进一步探讨X基因在乙型肝炎病毒(hepatitis B virus,HBV)感染者疾病转归中的作用。方法采用巢式PCR(nested-PCR),用型特异性引物对标本进行基因分型,针对基因分型成功的标本,采用巢氏PCR扩增X基因部分区段,对有特异性条带的扩增产物进行序列测定,测序结果用相关软件进行处理,翻译成蛋白质后与文献中已经公布的和疾病相关的变异位点进行比对并进行统计。结果测序成功的41人份标本翻译成氨基酸后,与文献中已报道的与疾病相关的X蛋白变异位点进行比较,发现17个样品的变异与肝细胞癌相关,9例X基因中的插入和缺失变异,会导致X蛋白整个读码框架的改变。结论本实验对HBV感染者的疾病转归进行了初步预测,通过追踪研究可对X基因在致病过程中所起的作用进行更为科学的调查和研究。

Analysis of Hepatitis B Virus X Gene in Some Sections and its Encoded Amino Acid Variation

Abstract:Objective To analyze X gene mutation in part of section and its encoded amino acid variation with serum or plasma sample infected with hepatitis B virus(HBV) and to discuss the function of X gene of HBV on forecasting disease progression.Methods The sequence specific primers were applied to genotype by the method of nested-PCR.For the samples of genotyping successfully.Nested-PCR was applied to amplify part of X gene segment with the DNA.The amplified liquid of X gene was sequenced.The relevant software was applied to analyse the results.Results 41 samples with X gene were successfully sequenced.After translating into amino acids,17 samples were found to be related to the variation of HCC through comparing the variable sites which had been reported in the literature.9 samples were found X gene mutation of insertion and deletion.This would result in the changes of X protein reading frame.Conclusions The experiment make a preliminary prediction of disease prognosis in people infected with HBV.The follow-up study is needed to patients.
