摘    要:目的:探讨丁酸钠对人绒毛膜癌JAR细胞生长抑制作用及其可能机制。方法:通过流式细胞仪定量分析细胞周期分布及细胞凋亡;并采用酶联免疫吸附法(ELISA)检测丁酸钠对Fas蛋白表达水平的影响。结果:丁酸钠对JAR细胞生长有抑制作用,呈剂量依赖关系(P<0.05);丁酸钠组细胞凋亡率明显高于对照组(P<0.01);随着丁酸钠浓度升高,Fas蛋白的表达水平逐步升高(P<0.05)。结论:丁酸钠可以抑制人绒毛膜癌JAR细胞的生长和诱导其凋亡,其机制可能与Fas蛋白的高表达有关。

Abstract:Objective: To study the growth-inhibiting effects of sodium butyrate on human chorionic carcinoma JAR cells and mechanism.Methods: The distribution of cell cycle and cell apoptosis were quantified using Flow cytometry(FCM);the effect of sodium butyrate on the expression of Fas protein in JAR cells was examined by ELISA.Results: The sodium butyrate had inhibiting effect on the growth of JAR cells and which presented dose-effect relation(P<0.05);the apoptosis rate of cell in sodium butyrate group was higher than that in control group(P<0.01);the expression rate of Fas protein increased following the increasing of sodium butyrate concentration(P<0.05).Conclusion: Sodium butyrate can inhibit the growth of human chorionic carcinoma JAR cells and induce their apoptosis,which may relate to the high expression of Fas protein.
