青蒿琥酯对胃癌耐药细胞SGC7901/mdr1阿霉素耐药性的影响

摘    要:目的以胃癌细胞SGC7901为对照,观察青蒿琥酯(Art)对胃癌耐药细胞SGC7901/mdr1的阿霉素耐药性的影响。方法四甲基偶氮唑蓝法检测Art耐药逆转性,将培养的SGC7901和SGC7901/mdr1细胞加入不同浓度的Art,确定Art逆转SGC7901/mdr1细胞阿霉素耐药性的浓度;加入不同终浓度的阿霉素并不断调整,直至对SGC7901/mdr1和SGC7901细胞的抑制范围均涵盖50%的抑制率,确定阿霉素的实验浓度;2种细胞均加入确定好的不同浓度阿霉素,SGC7901细胞设为亲本对照组,SGC7901/mdr1细胞分为2组,加入确定耐药逆转Art浓度的设为实验组,不加Art的设为Art阴性对照组,判断Art对耐药细胞SGC7901/mdr1的耐药逆转效果。结果 Art对SGC7901和SGC7901/mdr1细胞均有生长抑制作用,不同浓度Art之间两两比较差异均有统计学意义(P<0.05);Art对SGC7901/mdr1的50%抑制浓度高于SGC7901细胞(P<0.05);Art对SGC7901/mdr1和SGC7901细胞20%抑制浓度的差异无统计学意义(P>0.05),取二者的平均值110 mg·L-1作为Art对SGC7901/mdr1细胞的耐药逆转浓度。涵盖SGC7901/mdr1和SGC7901细胞50%抑制率的阿霉素质量浓度为3.125、6.250、12.500、25.000、50.000 mg·L-1。SGC7901/mdr1细胞实验组、Art阴性对照组和亲本对照组的细胞生长抑制率随阿霉素浓度的不断增加而增高,不同浓度阿霉素之间两两比较差异均有统计学意义(P<0.05);阿霉素对实验组细胞的50%抑制浓度低于Art阴性对照组(P<0.05),与亲本对照组比较差异无统计学意义(P>0.05)。Art对耐药细胞SGC7901/mdr1的耐药逆转倍数约为2.31倍,相对逆转率约为83.00%。结论 Art对SGC7901/mdr1有生长抑制作用;Art可以逆转耐药细胞SGC7901/mdr1对阿霉素的耐药性。

Abstract:Objective To observe the changes of artesunate( Art) on gastric cancer cells resistant to doxorubicin in SGC7901 /mdr1 with gastric cancer cells SGC7901 as control. Methods The resistance reversal was detected by tetramethyl even tetrazolium( MTT) assay: different concentrations of Art were added in the cultured cells SGC7901 and SGC7901 /mdr1,Art reversed determine the concentration of doxorubicin resistance SGC7901 /mdr1 cells; different concentrations of doxorubicin and constantly adjust the minimum and maximum final concentration,until on SGC7901 /mdr1 and SGC7901 cells were inhibited range covers 50% inhibition rate,determine the inhibition SGC7901 /mdr1 covering doxorubicin and SGC7901 cells by50% final concentration; two kinds of cells were added to determine the different concentrations of doxorubicin good,SGC7901cells set parental controls,SGC7901 /mdr1 cells were divided into two groups,the concentration of Art was determined by adding resistance reversal as test group,without Art was located to Art negative control group to determine Art on drug resistant cells SGC7901 /mdr1 reversal effect was determined. Results Art inhibited the growth of SGC7901 /mdr1 and SGC7901 cells,the differences among different concentrations of Art were statistically significant( P〈0. 05); IC50of Art on SGC7901 /mdr1was higher than SGC7901 cells( P〈0. 05),and there was no statistical significance between the IC20of Art on SGC7901 /mdrl and SGC7901( P〉0. 05),the average of 110 mg·L^-1 cells as Art on SGC7901 /mdr1 resistance reversal concentrations. The final concentration which covered the adriamycin inhibit SGC7901 / mdr1 and SGC7901 cells in 50% were 3. 125,6. 250,3. 125,6. 250,50. 000 mg·L^-1. The cell growth inhibition rate in experimental group,Art negative control group and parent controls increased with the increasing of concentration of adriamycin. There were significant differences of cell growth inhibition rate between the every two groups with different concentration of adriamycin(
