鸦胆子油乳联合顺铂对宫颈癌HeLa细胞的抑制作用及其机制

摘    要:目的研究鸦胆子油乳联合顺铂对宫颈癌HeLa细胞的增殖抑制与凋亡作用,探讨鸦胆子油乳联合顺铂的抗肿瘤作用及其机制。方法采用甲基噻唑基四唑法检测HeLa细胞生长抑制率;流式细胞仪检测各组药物对HeLa细胞凋亡的影响;免疫细胞化学法检测Survivin和Caspase-3蛋白表达水平。结果空白对照组、顺铂组、鸦胆子油乳组和鸦胆子油乳联合顺铂组HeLa细胞的凋亡率分别为(9.91±0.35)%、(13.65±0.18)%、(10.68±0.21)%、(15.56±0.37)%;顺铂组、鸦胆子油乳组、鸦胆子油乳联合顺铂组与空白对照组比较,差异均有统计学意义(P<0.01);鸦胆子油乳联合顺铂组与顺铂组和鸦胆子油乳组比较差异均有统计学意义(P<0.05,P<0.01)。空白对照组、鸦胆子油乳组、顺铂组和鸦胆子油乳联合顺铂组Survivin蛋白表达阳性细胞率分别为(84.22±2.96)%、(61.77±4.40)%、(29.47±3.74)%、(13.75±1.79)%,经两两比较差异均有统计学意义(P<0.01);Caspase-3蛋白表达阳性细胞率分别为(12.70±3.42)%、(38.32±2.76)%、(71.36±3.14)%、(91.31±2.23)%,经两两比较差异均有统计学意义(P<0.01)。结论鸦胆子油乳与顺铂联合对宫颈癌HeLa细胞增殖的抑制有协同作用;鸦胆子油乳联合顺铂诱导HeLa细胞凋亡可能与降低Survivin蛋白表达和增强Caspase-3蛋白表达有关。

Abstract:Objective To investigate the inhibition and apoptosis role of bruceolic oil emulsion combined with cisplatin on HeLa cells of cervical cancer,and explore its mechanism of action.Methods The proliferation inhibiting rate of HeLa cells was detected by methyl thiazolyl tetrazolium(MTT) assay; the flow cytometry was used to detect the effect of each drug on apoptosis of HeLa cells; the expression of Survivin and Caspase-3 protein were detected by immunocytochemistry.Results The apoptosis rate of HeLa cells in blank control group,cisplatin group,bruceolic oil emulsion group and bruceolic oil emulsion combined with cisplatin group was(9.91 ± 0.35) %,(13.65 ± 0.18) %,(10.68 ± 0.21) %,(15.56 ± 0.37) % respectively; there were statistic difference of apoptosis rate in each group(P〈 0.05,P〈 0.01).The expression of Survivin protein in blank control group,cisplatin group,bruceolic oil emulsion group and bruceolic oil emulsion combined with cisplatin group was(84.22 ±2.96) %,(61.77 ±4.40) %,(29.47 ±3.74) %,(13.75 ±1.79) % respectively and the expression of Caspase-3 protein in the four groups was(12.70 ± 3.42) %,(38.32 ± 2.76) %,(71.36 ± 3.14) %,(91.31 ± 2.23) % respectively;there were statistic differences of apoptosis rate in each group(P〈 0.01).Conclusion Bruceolic oil emulsion combined with cisplatin has synergistic effect on proliferation inhibition of HeLa cells.The mechanism of bruceolic oil emulsion combined with cisplatin induce the apoptosis of HeLa cells may be related with reducing the expression of Survivin protein and increasing the expression of Caspase-3 protein.
