西格列汀对2 型糖尿病鼠血清及不同组织视黄醇结合蛋白4的影响

摘    要:目的:研究西格列汀对2型糖尿病大鼠血清及不同组织视黄醇结合蛋白4(RBP4)表达水平的影响。方法将35只SD大鼠随机分为两组,即对照组(NC,n=10)和高脂组(HF,n=25)。12周后,HF组大鼠腹腔一次性注射链脲佐菌素35 mg/kg,成功建立2型糖尿病模型大鼠20只,并随机分为糖尿病对照组(DM组,n=10)和西格列汀干预组(SP组,n=10)。分别测定干预前后体质量(BW)、空腹血糖(FBG)、三酰甘油(TG)、胆固醇(TCH)、低密度脂蛋白(LDL-C)、空腹胰岛素(FINS)及血清RBP4水平,并计算胰岛素抵抗指数(HOMA-IR)和胰岛β细胞功能指数(HOMA-β)。Western Blot法检测大鼠不同组织RBP4蛋白表达水平。结果①与NC组相比,DM组、SP组BW、FBG、TG、TCH、LDL-C、FINS、HOMA-IR、RBP4升高,HOMA-β降低,差异有统计学意义(P<0.05);西格列汀干预后,SP组较DM组FBG、TG、TCH、LDL-C、FINS、HOMA-IR、RBP4降低,HOMA-β升高,差异有统计学意义(P<0.05)。②与NC组相比,DM组附睾脂肪、肌肉组织RBP4蛋白表达水平显著增加(P<0.05);与DM组相比,SP组附睾脂肪、肌肉组织RBP4蛋白表达水平显著降低(P<0.05)。结论2型糖尿病大鼠血清RBP4水平升高,附睾脂肪、肌肉组织RBP4蛋白表达水平增高。西格列汀能显著降低2型糖尿病大鼠血清RBP4水平及附睾脂肪、肌肉组织中RBP4蛋白表达水平。

Effects of sitagliptin on serum and different tissues retinol binding protein 4 expression levels in a rat model of type 2 diabetes mellitus

Abstract:Objective To evaluate the effects of sitagliptin on metabolic parameters,serum retinol binding protein 4 (RBP4)lev-els,liver,epididymal adipose tissue and skeletal muscle RBP4 expression levels in a rat model of type 2 diabetes mellitus (T2DM). Meth-ods Tirty-five Sprague Dawley rats were randomly divided into normal diet group (NC group)of 10 rats and high fat diet group (HF group) of the remaining 25. After being fed on a high fat diet for 12 weeks,the HF group was given the peritoneal injection of a low dose of strepto-zotocin 35 mg/kg,and a rat model of T2DM (n=20)was established and randomly divided into two groups:diabetic control group (DM group,n=10),sitagliptin group (SP group,n=10). Body weight (BW),fasting blood glucose (FBG),triglyceride (TG),total choles-terol (TCH),low-density lipoprotein (LDL-C),fasting insulin (FINS),serum RBP4 were measured respectively before and after the 6 weeks of treatment,and homeostasis model assessment-insulin resistance index (HOMA-IR),homeostasis model assessment β-cell function index (HOMA-β)were calculated. The liver,epididymal adipose tissue and skeletal muscle RBP4 protein expression levels were detected by Western blot. Results ①The levels of BW,FBG,TG,TCH,LDL-C,FINS,HOMA-IR and serum RBP4 in DM group and SP group in-creased while the levels of HOMA-β decreased compared with those of NC group with a statistical significance (P<0. 05 ). After treatment with sitagliptin,the levels of FBG,TG,TCH,LDL-C,FINS,HOMA-IR and serum RBP4 of SP group decreased while the levels of HOMA-β increased compared with those of DM group with a statistical significance (P<0. 05 ). ② Epididymal adipose tissue and skeletal muscle RBP4 protein expression levels in DM group increased compared with those in NC group,and epididymal adipose tissue and skeletal muscle RBP4 protein expression levels in SP group decreased compared with those in DM group. Conclusion Serum,epididymal adipose tissue and skeletal muscle RBP4 protein levels are elevated in a rat model of T2DM. Sitagliptin can lower serum,epididymal adipose tissue and skeletal muscle RBP4 protein levels in a rat model of T2DM.
