淫羊藿总黄酮的提取工艺及含量分布

摘    要:目的研究淫羊藿总黄酮的提取工艺及产地不同的淫羊藿植株总黄酮在各部位的含量分布。方法用乙醇和水回流提取淫羊藿总黄酮;选取湖北恩施及福建三明淫羊藿为原料,做单因素实验,再以淫羊藿总黄酮提取率为考察指标,作正交实验设计;采用紫外分光光度法测定淫羊藿总黄酮含量。结果以乙醇和水作为提取剂的提取率分别为70.5%、31.1%;恩施与三明所产的箭叶淫羊藿其淫羊藿总黄酮含量分别为:叶3.24%,1.02%;茎0.91%,0.14%;根2.47%,0.61%。结论湖北恩施所产的箭叶淫羊藿比福建三明所产的箭叶淫羊藿中总黄酮含量高;乙醇作提取剂的提取率明显高于水,影响淫羊藿总黄酮提取效果的主次因素为:溶剂浓度>回流时间>提取次数>液固比。淫羊藿总黄酮的优化提取工艺为:液固比为12∶1,以60%乙醇提取3次,1.5 h/次,优化工艺的总黄酮提取率达77.5%。

Abstract:Objective To study an extraction Technology for total flavone and to determine the content distribution in Herba Epimedium.Methods The extraction rate was compared between ethanol extract and water extract.The optimum extractive conditions were investigated by orthogonal design and one-factor experiment.Results The extraction rates with ethanol and water as extractants were(65.2)%,26.6%,respectively.The contents of Epimedium flavonoids from Enshi in root,stalk and leaf were(3.24)%,0.91% and(2.43)%,respectively,while the ones from Sanming were 1.02%,0.14%,0.61%,respectively.Conclusion The factors which influence extraction rate are sorted as follows: A>C>D>B,where A,B,C and D stands for the concentration of ethanol,the dosage of ethanol,extract time and times of leaching,respectively.The optimum technology of extraction is as follows: 60% ethanol,twelve-fold leachant,3 times extraction and 1.5 hours.The extraction rate is up to 77.5%.
