大鼠胶原诱导关节炎模型的建立及评价

目的建立Ⅱ型胶原诱导关节炎（CIA）大鼠模型及评价方法。方法大鼠皮内注射鸡Ⅱ型胶原（CⅡ）与完全弗氏佐剂及不完全弗氏佐剂建立模型,监测动物体重、后足容积、关节炎指数（AI）以及热刺激缩足反射潜伏期（PWTL）和机械痛阈（MWT）,免疫后第四十二天行影像学检查并取材进行组织病理学检查;酶联免疫吸附法（ELISA）检测动物血清抗CⅡ抗体、IL-1β及TNF-α水平。结果 CIA大鼠体重增长缓慢,后足红肿明显,后足容积显著高于对照组,AI呈上升趋势,后足PWTL及MWT均显著降低,影像学检查发现关节腔狭窄,关节结构模糊,组织病理学检查可观察到明显炎症细胞浸润、关节结构的破坏及滑膜增生,ELISA结果显示CIA大鼠血清抗CⅡ抗体、IL-1β及TNF-α水平显著升高。结论本文成功建立了CIA大鼠模型及模型评价方法。

Establishment and Evaluation the Arthritis Model Induced by Collagen in Rats

OBJECTIVE To establish the rat model of collagen-induced arthritis （CIA） and the method of evaluation. METHODS CIA was induced in male wistar rats by injecting subcutaneously with chicken type H collagen （C Ⅱ ） in Freund＇ s complete adjuvant （CFA） and Freund＇ s incomplete adjuvant（IFA）. Arthritis was evaluated by body weight, hind paw volume, arthritis index （AI）, pain thresholds including paw withdraw thermal latency （PWTL） and mechanical withdrawal threshold （MWT）, radiographic and histological examination. Levels of interleukin-1β （IL-1β）,tumor necrosis factor-α （TNF-α） and anti-C Ⅱ antibody in serum were determined by Enzymelinked immunosorbent assay （ELISA）. RESULTS Compared to normal control group,the CIA rats showed a slower increase of body weight, their hind paws appeared red and swelling, hind paw volume was significantly increased, the scores of AI increased, both PWTL and MWT were significantly decreased. X-ray examination showed that the joint space was narrow and the joint structure was broken, and histological pathology of ankle joints showed that the synovium of CIA rats were hyperplasia, and inflammatory cells infiltrated into synovium. Significantly increased levels of anti-C Ⅱ antibody, IL-1β and TNF-α were observed by ELISA. CONCLUSION This study has successfully established CIA rat model and the method of evaluation.
