血液病患者血及骨髓涂片中微小病毒B19的检测

目的 :用聚合酶链式反应 (PCR)技术检测白血病患者骨髓及血涂片中微小病毒 B19- DNA,从而了解白血病患者 B19的感染情况。方法 :收集临床已确诊的白血病患者的骨髓及血涂片标本 ,其中 ,骨髓涂片标本 43例 (急性淋巴细胞白血病 17例 ,急性非淋巴细胞白血病 18例 ,混合白血病 2例 ,慢性粒细胞性白血病 6例 ) ,血涂片 30例 (急性淋巴细胞白血病 12例 ,急性非淋巴细胞白血病 14例 ,慢性白血病 4例 ) ,正常体检人员血涂片 30例。用本文建立的PCR及限制性内切酶酶切检测的方法检测所收集标本的 B19- DNA进行病例对照研究。结果 :骨髓涂片和血涂片之间 B19的检出率无统计学差异 (P>0 .0 5 ) ,但是二者与正常对照组相比均有统计学差异 ,分别为 P<0 .0 0 5和 P=0 .0 5。急性淋巴细胞白血病、急性非淋巴细胞白血病和慢性白血病 B19的检出率之间无统计学差异 (P>0 .0 5 )。结论 :研究显示白血病患者由于化疗及免疫功能低下 ,易感染 B19病毒 ,病毒感染后易诱发骨髓造血功能低下。

Detection of human parvovirus B19 on blood and bone marrow samples among adult with leukemia

Objective:To detect B19-DNA on blood and bone marrow samples among adult with leukemia by using polymerase chain reaction(PCR) stated the parvovirus B19 infection in adult with leukemia.Methods:We conducted a case-control study to investigate the infection of B19-DNA on blood and bone marrow samples of 43 cases with acute and chronic leukemia,30 cases of health person with PCR technique.Results:Comparing the position rate in bone marrow samples with that in blood samples in B19-DNA,there was no statistical difference(P>0.05),but both of them were significantly higher than that in health person(P>0.005,P=0.05).Comparing the position rate among acute non-lymploblastic leukemia,acute lymphoblastic leukemia and chronic leukemia,there no statistical diference(P>0.05).Conclution:These result showed that persons with leukemia who received chemotherapy and were immunosuppressed easily underwent B19 infection,inducing bone marrow failure.
