补肾健脾药物血清对体外培养的成骨细胞增殖与分化的影响

目的观察补肾健脾药物血清对新生大鼠颅骨成骨细胞增殖和分化的影响.方法用多次酶消化法分离培养新生大鼠颅盖骨成骨细胞,加入含不同药物浓度的补肾健脾药物血清条件培养液共同培养;改良Gomori钙钴法染色鉴定,MTT法测定细胞增殖率,对硝基苯酚法检测碱性磷酸酶(ALP)活性.骨疏康和尼尔雌醇作为阳性对照药物.结果补肾健脾中药组成骨细胞的增殖、ALP活性均显著高于空白对照组(P＜0.05),与骨疏康和尼尔雌醇组比较有显著性差异.结论补肾健脾中药可通过增加成骨细胞的增殖与分化而促进骨形成.

Experimental Research of the Serum Contained Tonic-kidney and Benefiting Spleen Herbs Function to the Proliferation and Differentiation of the Vitro-Cultured Osteoblasts

Objective To observe the effects of the serum contained tonic-kidney and benefiting-spleen herbs to the proliferation and differentiation of osteoblasts of newly born rats' cranium. Method We separated the osteoblasts of newly born rats' cranium and cultured them in vitro with the method of twice enzyme-digestion. And supplying the osteoblasts drug-contained serum which has different concentration. We identified the cells by means of Gomori staining method and detected the proliferation rate of the cells by MTT. The activity of the alkaline phosphatase of the cells was examined by the method of para-nitryl phenol. And we also chose the Gushukang and nilestriol as the control drugs. Results The Chinese herbs which have the function of tonifying the kidney and benefiting the spleen can stimulate the proliferation of the osteoblasts and the activity of the alkaline phosphatase, which are dramatically better than the blank-contrast group. To be compared with the drug- control groups, it has no dramatic significance in statistics. Conclusion The Chinese herbs which have the function of tonifying the kidney and benefiting the spleen can accelerate the bone formation by the way of stimulating the proliferation and differentiation of the osteoblasts.
