Calbindin-D28k mRNA在大鼠三叉神经节和背根节初级传入神经元中的表达

摘    要:用原位杂交组织化学技术,用同位素标记的寡核苷酸探针,对Calbindin-D28kmRNA在大鼠三叉神经节和背根节初级传入神经元中的表达进行了观察.结果发现:在大鼠三叉神经节中,约16.1%的神经元为CalbindinD28k阳性神经元,而在背根神经节中,阳性神经元占节细胞总数的20.6%.这两个神经节中的阳性神经元主要集中在大型细胞(8.1%和12.3%)及小型细胞(4.2%和6.8%).从而提示Calbindin-D28k可作为上述两种神经细胞的标志物,在此类初级传入神经元的某些生理功能中可能有重要作用.

Abstract:Calbindin- D28k (CaB), a member of large family of calcium-binding proteins (CaBP), is expressed abundantly in thenervous system. But the presence of CaB has been received very little attention in the trigeminal ganglia(TG) and dorsal rootganglia(DRG). Accordingly, Using in situ hybridization histochemistry with specific oligonucleotide probe for CaB mRNA,we examined the expression of CaB gene in the TG and DRG of rat. Spragne-Dawley rats were used. The TG and DRG wereremoved by decapitation and frozen in dry ice quickly. The sections were cut on a cryostat microtome and thaw-mounted ongelatin-coated slides. These sections were incubated with the probes labeled at 3'end using α-35 S]dATP, washed at highstringency and examined by emulsion autoradiography. In rat TG, about 16. 1% of the neurons were labled with CaB.Whereas, the percentage of CaB-positive neurons in DRG was 20. 6 %. The positive signals were mainly confined to two subpopulations of neurons: large cells and small cells. Therefore, CaB may act as a cell marker of these two subpopulations ofneurons in TG and DRG. Furthermore, CaB may participate and medulate many important calcium-dependent processes inprimary sensory neurons.
