RUNX3蛋白在结直肠癌中的表达及意义

通过高通量组织芯片技术，制作石蜡组织微阵列，运用免疫组织化学ABC方法，检测RUNX3蛋白在癌旁及肿瘤组织中的表达特点，分析其与结直肠癌各病理特征之间的关系，探求该基因在结直肠癌中表达的生物学意义，以期为结直肠癌的早期诊断、预后评价和基因治疗指标的筛选提供有价值的实验依据。结直肠癌和癌旁组织均来自未经化放疗的诊断明确的结直肠癌手术切除标本62例及相应癌旁组织21例。采用石蜡组织微阵列技术，运用HE染色明确病理诊断，应用免疫组织化学染色方法，对RUNX3在非癌组织及结直肠癌中的表达情况进行检测与统计。实验数据采用SPSS11．5软件包进行统计学差异分析。结果显示：（1）在21例癌旁相对正常组织中和62例结直肠癌组织中，RUNX3阳性表达率分别为95．2％（20／21）和69．4％（43／62），癌旁相对正常组织中RUNX3蛋白的阳性染色率高于结直肠癌组织（P=0．017）。在62例肿瘤中16例结肠肿瘤和46例直肠标本中RUNX3阳性率分别为（12／16，75．0％），（31／46，67．4％），两者无统计学差异（P=0．573）。（2）RUNX3的表达与肿瘤各病理指标之间关系：在45例溃疡型样本和17例隆起型样本中，RUNX3表达阳性率分别为62．2％（28／46）和88．2％（15／17），二者之间有统计学差异（P=0．049）。在25例无淋巴结转移样本和37例有淋巴结转移样本中，RUNX3表达阳性率分别为84．0％和59．5％，二者之间有统计学差异（P=0．041）。RUNX3的表达在不同性别、年龄、分化程度、病变大小及不同浸润深度间无统计学差异。结果表明：（1）结直肠癌组织中RUNX3蛋白表达较癌旁黏膜组织呈现显著降低下调趋势，提示RUNX3蛋白表达下调可能与直肠癌的发生、发展有关。（2）RUNX3基因表达无年龄、性别、分化程度、病变大小、浸润深度间差异，但与肿

Expression and Significance of RUNX3 Protein in Colorectal Cancer

High-flux tissue-chip was used to make paraffin tissue microarrays;immunohistochemlstry ABC method was used to detect the expression of RUNX3 in tumor and paraneoplastic tissue and to analyze its relationship with various pathological features of colorectal cancer, so that the biological significance of RUNX3 expression in colorectal cancer could be revealed, so as to explore valuable experimental basis for early diagnosis,prognosis and screening of gene therapy indexes of colorectal cancer is taken. Tumor and paraneoplastic tissue were from 62 cases of surgical specimens and 21 cases of the relevant paraneoplastic specimens in confirmed colorectal cancer neither receiving chemotherapy or radiotherapy. Paraffin tissue microarrays and HE staining were used to confirm the pathological diagnosis. Immunohistochemical staining method was used to detect RUNX3 expression in non-cancerous tissue and in colorectal cancer and to make a statistics analysis. Experimental data were analyzed for statistical difference with SPSS11. 5 soft ware package. The results showed that： 1）the positive expression rate of RUNX3 was 95.2% （20/21）and 69.4 % （43/62）respectively in 21 cases of paraneoplastic relatively normal tissue specimens and 62 cases of colorectal cancer tissue specimens. The positive staining rate of RUNX3 was higher in paraneoplastic relatively normal tissue than in cancer tissue（ P =0. 017）.
