无毛小鼠接种申克孢子丝菌检测抗申克孢子丝菌细胞外蛋白酶抗体研究

摘    要:通过申克孢子丝菌接种无毛小鼠(Hairlesmice)来检测两种细胞外蛋白酶的血清抗体。在小鼠皮内注射申克孢子丝菌并形成结节,镜下示真菌消失,并在每周通过酶联免疫吸附试验(EIA)检测蛋白酶Ⅰ和Ⅱ的血清抗体。结果显示抗体滴度与实验小鼠孢子丝菌病肉眼和镜下观察的变化趋势相一致。本试验证明了申克孢子丝菌在体内能产生蛋白酶Ⅰ和Ⅱ,而且蛋白酶的抗体可用于孢子丝菌病的血清学诊断。

The level of antibody against extracellular Proteinase of Sporothrix schenckii in S.schenckii inoculated hairless Mice.
Abstract:Sporothrix schenckii produces two extracellular proteinases,named proteinase I and II.Studies suggested an important role for these proteinases in terms of fungal invasion and growth.Subsequent to an intracutaneous injection of S.schenckii to the moues skin,nodules spontaneously formed and disappeared for a period of 4 weeks.Histopathological examination results were in accordance with the microscopic observation.Serum antibody titers against purified proteinases I and II were measured weekly,Using EIA.As a result,the time course of the antibody titers to both I and II were parallel to that of macroscoopic and microscopic observations in an experimental mouse sporotrichosis model.These results suggest that S.schenckii produces both proteinases I and II in vivo.Moreover,the detection of antibodies against these proteinases can contribute to a serodiagnosis of sporotrichosis.
