摘    要:目的:提取漏芦多糖,分析其单糖组分并测定其质量分数,为以后漏芦多糖研究提供参考。方法:水提醇沉法提取粗多糖,Sevag法除蛋白,高效阴离子交换色谱-脉冲安培检测( HPAEC-PAD )法测定其单糖组成,苯酚-硫酸法测定多糖质量分数。结果:漏芦多糖的单糖组成为葡萄糖、阿拉伯糖和果糖,其摩尔比为1∶1.61∶2.21。以单糖混合物为对照品测得漏芦多糖中总糖的质量分数为 95.78%。结论:HPAEC-PAD法具有样品不用衍生、灵敏度高、精密度好等优点,适合多糖类样品中单糖的分析;苯酚-硫酸法实验操作简便,准确可靠,可用于多糖含量测定。

Abstract:OBJECTIVE: To analyze the monosaccharide composition in the polysaccharides from Rhaponticum uniforum, determine the content of monosaccharide, and provide some references for further research. METHOD: The monosaccharide composition was determined by high performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD). Phenol-sulfuric acid method was used for the determination of the content of polysaccharide. RESULT: The monosaccharides composition in polysaccharides from R. uniforum are glucose, arabonose and fructose. Their molar ratios are 1 : 1.61 : 2.21. The content of polysaccharide is 95.78%, taking the mixture of monosaccharide compositions as reference substances. CONCLUSION: HPAEC-PAD can be used to analyze the monosaccharide composition in the polysaccharide with high precision, and the method of phenol-sulfuric acid is simple, convenient and reliable.
