~(60)Coγ-射线对冻干血浆中病毒灭活效果的研究

摘    要:为提高冻干血浆临床应用的安全性,采用细胞感染法对人工污染病毒的新鲜冰冻血浆经60Co辐照灭活处理的效果进行了检测,同时用凝胶电泳和凝血因子分析仪检测60Co照射对冰冻血浆生物活性物质的影响。结果,60Coγ-射线经30 kGy的辐照剂量能完全灭活冻干血浆中的指示病毒;照射后的血浆白蛋白等成分含量略有下降,凝血因子活性减少了30%~40%。结论,60Coγ-射线辐照能有效灭活冻干血浆中的有包膜病毒和无包膜病毒,但对血浆蛋白活性有一定影响。

Abstract:In order to improve the safety of clinical use of lyophilized plasma,cell infection method was used to examine the efficacy of ~(60)Co irradiation sterilization treatment of fresh lyophilized plasma artificially contaminated by virus.At the same time,gel electrophoresis and coagulation factor analyser were used to examine the influence of ~(60)Co irradiation on biologically active substances in lyophilized plasma.Results: 30 kGy ~(60)Co γ-irradiation could completely inactivate the indicator virus in lyophilized plasma.The contents of plasma components such as albumin decreased slightly and the activities of the coagulation factors decreased by 30%~40% after irradiation.Conclusion: ~(60)Co γ-irradiation can effectively inactivate the viruses with and without envelope in lyophilized plasma,but had certain influence on activities of plasma proteins.
