| 摘 要： | 目的观察二硫化二砷（As2S2）对人肝癌细胞株（BEL-7402）凋亡和坏死的影响。方法人肝癌细胞株BEL-7402经不同浓度A82s2处理后，采用MTT法进行细胞增殖抑制率的测定，台盼蓝排染法观察As2S2对肝癌细胞的细胞毒作用，通过流式细胞仪定量检测细胞凋亡和坏死的情况，同时检测凋亡相关蛋白Fas和Apo2．7的表达。结果As2S2对细胞有明显细胞毒作用，并且可诱导细胞凋亡的产生，上调Apo2．7和Fas蛋白的表达。结论A22S2具有促肝癌细胞凋亡和坏死的作用。

| Abstract: | Abstract： Objective To study the effect of Ass S2 on apoptosis and necrosis of human hepatoma cell line in vitro. Methods BEL- T402 cell line was treated with Ass S2 at different concentration. The proliferation inhibition was detected by M＇IT assay. Cytotoxity was observed by cell counting. Neeresis was estimatedby Trypan blue staining. Thecell apoptosis and necrosis were detected by cytometer with Annexin-V/PI. The expressions of Apo2.7 and Fas protein were also detected by cytometer. Results The results showed that high dose of As2S2 had cytotoxity on BEL-7402. In addition, cell apoptosis was detected by cytometer, and the expressions of Fas and Apo2.7 were up-regulated. Conclusion Ass $2 can induce apoptosis and necrosis of hepatoma cells. |
