丙泊酚对大鼠的空间学习记忆和海马NMDA受体NR2A亚基表达的影响

摘    要:目的观察丙泊酚单次麻醉对成年大鼠空间学习记忆能力的影响以及海马NMDA受体NR2A亚基的变化。方法 30只SD大鼠随机分成丙泊酚组(n=15)和对照组(n=15)。给予丙泊酚50mg/kg腹腔注射作为诱导剂量,待翻正反应消失后在腹腔追加50mg/kg丙泊酚加深麻醉。对照组不予任何处理。待翻正反应恢复12h后应用Morris水迷宫评价丙泊酚对于大鼠空间学习记忆的影响。同时通过免疫荧光技术检测海马NMDA受体NR2A亚基的表达变化。结果丙泊酚组平均学习潜伏期较对照组显著缩短(P〈0.05),靶象限活动时间百分比明显延长(P〈0.05),丙泊酚组NMDA受体NR2A亚基的表达增加。结论丙泊酚单次麻醉后成年大鼠的空间学习记忆能力提高,这种学习记忆能力的提高可能和NMDA受体NR2A亚基的表达上调相关。

Abstract:Objective To investigate the effect of propofol on the spatial performance in Morris water maze(MWM)and the changes in hippocampal NMDA receptor NR2A subunit by immunofluorescence in rats.Methods Thirty Sprague-Dawley rats were randomly divided into propofol group and control group.An intraperitoneal injection of propofol(50 mg/kg)was performed in the propofol group.After righting reflex of rats were lost,propofol(50 mg/kg)was administered intraperitoneally again in order to maintain anesthesia.The control group was blank.Twelve hours later,rats were subjected to MWM so that their spatial learning and memory could be evaluated.Meanwhile the changes in hippocampal NMDA receptor NR2A subunit expression were measured by immunofluorescence.Results The escape training latencyin of the propofol group was shorter than that of the control group(P〈0.05),the time spent on target quadrant in probe test was longer in the propofol group than in the control group(P〈0.05).Hippocampal NMDA receptor NR2A subunit expression was increased in the propofol group.Conclusion Spatial learning and memory is improved and expression of hippocampal NMDA receptor NR2A subunit obviously is increased in rats after single anesthesia by propofol.
