盆腔器官脱垂患者骶韧带成纤维细胞粘弹性研究

摘    要:目的探讨盆腔器官脱垂(pelvic organ prolapse,POP)患者骶韧带成纤维细胞粘弹性变化。方法选取2009年12月至2010年6月河北医科大学第二医院妇产科POP患者11例为观察组,同期非POP患者13例为对照组,取骶韧带进行成纤维细胞的原代培养。光镜观察与免疫细胞化学染色法进行成纤维细胞鉴定,取1代的成纤维细胞(实验组观察细胞数为18,对照组观察细胞数为22),采用微管吸吮技术表征骶韧带成纤维细胞的粘弹性特性。结果培养的细胞呈梭形,表达波形蛋白(vimentin),POP患者骶韧带成纤维细胞粘弹特性明显低于正常对照组,病例组骶韧带成纤维细胞平衡模量[E∞=(124.62±20.22)Pa]、瞬间模量[E0=(429.25±105.89)Pa]、黏性系数[μ=(718.53±159.08)Pa.s]均显著低于对照组[E∞=(170.17±24.65)Pa]、[E0=(495.26±69.39)Pa、μ=(845.35±155.34)Pa.s],P<0.05。结论 POP患者成纤维细胞骨架结构和机能状态的改变可能是POP发生的关键环节。
