摘    要:目的:建立一种具有较高GSH含量和GST活力的肝细胞系。方法:将HepG2细胞在0.1,0.3,0.5mg/L浓度的CDDP间歇作用下传代12个月后得到三种稳定的CDDP耐药细胞系HepG2/CDDP0.1,HepG2/CDDP0.3,HepG2/CDDP0.5细胞的GSH含量和GST活力,结果用显著性t检验方法作统计学处理。结果:诱导刺激后的子代HepG2/CDDP0.1,HepG2/CDDP0

Abstract:Objective :To develop a kind of HepG2 variant with high GSH content and GST activity.Methods: HepG2 cells were pulse treated with 0.1, 0.3, 0.5 mg/L CDDP for 1h every week, respectively, Twelve months later , three CDDP-resistant cell lines (HepG2/CDDP0.1, HepG2/CDDP0.3, HepG2/CDDP0.5 ) were obtained. GSH content and GST activity were measured. The results were statistically analyzed by the method of t-test. Results:There were two-fold elevated of GSH and GST exist In HepG2/CDDP0.1, HepG2/CDDP0.3, HepG2/CDDP0.5 comparing to the parental HepG2. And there were significant discrepancies of GSH contains between HepG2 and HepG2/CDDP 0.3, HepG2/CDDP 0.5. Conclusion: All evidence suggest that GSH and GST can be induceed under the treatment of CDDP.
