| 摘 要： | 目的：建立一种具有较高ＧＳＨ含量和ＧＳＴ活力的肝细胞系。方法：将ＨｅｐＧ２细胞在０．１，０．３，０．５ｍｇ／Ｌ浓度的ＣＤＤＰ间歇作用下传代１２个月后得到三种稳定的ＣＤＤＰ耐药细胞系ＨｅｐＧ２／ＣＤＤＰ０．１，ＨｅｐＧ２／ＣＤＤＰ０．３，ＨｅｐＧ２／ＣＤＤＰ０．５细胞的ＧＳＨ含量和ＧＳＴ活力，结果用显著性ｔ检验方法作统计学处理。结果：诱导刺激后的子代ＨｅｐＧ２／ＣＤＤＰ０．１，ＨｅｐＧ２／ＣＤＤＰ０

| Abstract: | Objective :To develop a kind of HepG2 variant with high GSH content and GST activity.Methods: HepG2 cells were pulse treated with 0.1, 0.3, 0.5 mg/L CDDP for 1h every week, respectively, Twelve months later , three CDDP-resistant cell lines (HepG2/CDDP0.1, HepG2/CDDP0.3, HepG2/CDDP0.5 ) were obtained. GSH content and GST activity were measured. The results were statistically analyzed by the method of t-test. Results:There were two-fold elevated of GSH and GST exist In HepG2/CDDP0.1, HepG2/CDDP0.3, HepG2/CDDP0.5 comparing to the parental HepG2. And there were significant discrepancies of GSH contains between HepG2 and HepG2/CDDP 0.3, HepG2/CDDP 0.5. Conclusion: All evidence suggest that GSH and GST can be induceed under the treatment of CDDP. |
