摘    要:目的证明肿瘤抑制因子Tob1是否增加人类乳癌细胞系MDA-MB-231对电离辐射的敏感性,并初步探讨Tob1发挥辐射增敏作用的机制。方法采用集落形成实验探讨重组腺病毒介导的Tob1对MDA-MB-231辐射敏感性的影响,并通过Western Blot法检测由Tob1诱导的受γ线照射后乳癌细胞中相关蛋白表达水平的改变。结果重组腺病毒介导的Tob1高水平表达可显著增加乳癌细胞MDA-MB-231对电离辐射的敏感性(P<0.05),并可引起受照射后乳癌细胞中与凋亡及DNA损伤修复相关蛋白的表达水平改变。结论肿瘤抑制因子Tob1可能通过对细胞凋亡及DNA损伤修复相关蛋白的表达调控,增加乳癌细胞对电离辐射的敏感性。

Abstract:Objective To investigate the effects of the tumor suppressor Tob1 on radiosensitivity in a human breast cancer cell line MDA-MB-231.Methods Cell survival was determined by clonogenic assay by using adenovirus expression system(Ad5),and protein expression was analyzed by Western blot assay.Results It was demonstrated that pre-irradiation treatment with Ad5-Tob1 significantly increased the radiosensitivity of MDA-MB-231.Furthermore,Ad5-Tob1-mediated radiosensitivity was related to up-regulation of pro-apoptotic protein BAX and to down-regulation of several DNA DBS repair proteins.Conclusion Tob1 may be a radiosensitizer of breast cancer via increasing apoptosis and suppressing DNA repair.
