补肾填精复方对WI38细胞周期及相关基因表达的影响

摘    要:目的:观察补肾填精中药复方对衰老细胞周期及相关基因表达的影响.方法:选用衰老细胞模型WI38常规培养至33代,加入补肾填精复方含药血清作用至40代,并以流式细胞仪、RT-PCR、wester-blot等方法检测补肾填精含药血清对衰老细胞周期及相关基因表达影响.结果:33代后传代细胞出现明显衰老特征;衰老细胞阻滞于G1期;补肾填精含药血清可以延缓WI38细胞衰老,促进细胞进入细胞周期;补肾填精含药血清可降低细胞衰老相关基因P53、P21、P16转录与表达水平.结论:补肾填精中药复方可以影响细胞周期,减缓细胞衰老,其作用的环节可能与降低细胞衰老相关基因P53、P21、P16表达水平相关.

Abstract:Objective:To observe the effects of Chinese herbal Bu-Shen-Tian-Jing on cell cycle and the expression of senescence related gene of aging cell.Methods:WI38 cell was incubated with Bu-Shen-Tian-Jing serum from 33 propagation to 40 propagation.The effects of BuShen-Tian-Jing on process of cell cycle and the expression of senescence related gene of WI38 was measured by using flow cytometer,RT-PCR and western blot.Results: WI38 cell was aged and arrested at G1 phase from 33 propagation.Bu-Shen-Tian-Jing can delay the age process of WI38 cell,and stimulate WI38 cell enter cell cycle,and down-regulate the expression of P53,P21 and P16 gene.Conclusions: Bu-Shen-Tian-Jing can delay the age process of WI38 cell,and promote cell enter cell cycle,and those effects was associated with down-regulate the expression of P53,P21 and P16 gene.
