雷公藤单体T4对IL-1刺激引起的类风湿患者关节滑膜成纤维细胞增殖和产生IL-6的影响

摘    要:目的 探讨雷公藤单体 T4对经 IL- 1刺激的体外培养的类风湿关节炎 (RA)患者的关节滑膜成纤维细胞增殖和产生 IL- 6的影响。方法 关节滑膜组织来自行关节置换术或滑膜切除术的类风湿患者。滑膜组织经体外消化使细胞分散后 ,传代培养。以培养 3代后的滑膜成纤维细胞为对象 ,先给予 IL- 1刺激 ,再加入雷公藤 T4单体。结果  T4可抑制由 IL- 1刺激的滑膜成纤维增殖 ,但不影响 IL- 6的产生。结论  T4的上述作用可能是其治疗RA的机制之一 ,本研究结果为 T4早日应用于临床治疗 RA提供了一定的理论依据

Abstract:Objective Tripterygium Wilfordii Hook multiglycosides T 2 has been wildly used in China in treatment of RA. T 4 was abstracted from T 2 and was reported much more efficient in antiinflammatory and immune suppression than T 2.This study was to investigate the effect Tripterygium Wilfordii Hook T 4 monomer on proliferation and interlukin6 production of synovial fibroblasts of patients with rheumatoid arthritis. Methods Synovium was obtained from patients with rheumatoid arthritis undergoing synovectomies or joint replacement. Cultures of synovial fibroblasts were established.After 3 generations, cultured synovial fibroblasts were stimulated with IL1. Then 15 ng/ml, 5 ng/ml and 15 ng/ml T 4 were added, and synovial fibroblasts were cultured in the presence of T 4 for 48 hours. Cell proliferation was assayed using MTT method. IL6 level of supernatant was measured by ELISA. Results Proliferation of synovial fibroblasts was inhibited by T 4.The proliferation inhibition effect of T 4 was dose dependent and inhibition rate was 518%, 1095% and 2137%, respectively. And T 4 had no effect on IL6 production by IL1 stimulated synovial fibroblasts. Conclusions T 4 might control the disease activity of RA by inhibiting the proliferation of synoviocyte. And T 4 might not influence the concentration of IL6 in synovial fluid, as a central effect, since IL6 has protective effect on articular cartilage.
