神经节苷脂GM1对神经生长因子诱导PC12细胞分化的影响

目的 研究神经节苷脂GM1是否介导神经生长因子（NGF）诱导的PC12细胞的分化。方法 采用MTT法、葡萄神经酰胺合成抑制剂（D－PDMP）分析神经节苷脂GM1对NGF诱导的PC12细胞分化的影响。结果 神经节苷脂GM1对PC12细胞的生长无明显的影响，而能够促进NGF对PC12细胞的生长抑制作用；NGF能诱导PC12细胞分化，但NGF无法诱导神经节苷脂缺乏型PC12细胞的分化；在神经节苷脂缺乏型PC12细胞中添加神经节苷脂GM1后，细胞恢复对NGF的反应性。结论 神经节苷脂GM1在NGF诱导的PC12细胞分化过程中发挥重要作用。

Effects of Ganglioside GM1 on the Differentiation of PC12 Cells Induced by Nerve Growth Factor

Objective To determine if ganglioside GM1 affects the differentiation of PC12 cells induced by nerve growth factor(NGF). Methods The effect of GM1 on the differentiation of PC12 cells induced by NGF was investigated by MTT assay, inhibitor of glucosylceramide synthase D|threol|phenyl|2|decanoylamine|3|morpholinopropanol(D|PDMP). Results Ganglioside GM1 had no effect on the growth of PC12 cells, but it could enhance the inhibitory effect of NGF on the growth of PC12 cells. NGF could induce the differentiation of PC12 cells, but it didn't have the same effect on the PC12 cells lack of gangliosides. While with GM1, PC12 cells lack of gangliosides could response to NGF again. {Conclusion} Ganglioside GM1 plays an important role in the differentiation of PC12 cells induced by NGF.;
