血管紧张素原和内皮型一氧化氮合酶基因多态性与高血压病的相关性

摘    要:目的探讨血管紧张素原(AGT)M235T基因、内皮型一氧化氮合酶(eNOS)G894T基因多态性与原发性高血压(EH)的关系。方法采用基因芯片技术检测224例(EH组和对照组)AGT和eNOS基因的多态性,并比较两组基因型和等位基因频率。结果EH组AGT TT基因型携带者频率(74.19%)显著高于对照组(55.00%),χ2=9.04,P<0.01。EH组eNOS-GT+TT基因型频率(12.90%)与对照组(15.00%)相比差异无显著性(P>0.05)。结论AGT-TT基因多态性可能是中国人高血压病的独立危险因素,eNOS基因G894T多态性可能不是高血压病的独立危险因素。基因芯片技术为研究多种易感基因与高血压病的相关性提供了一项高效、敏感的方法。

Abstract:Objective To evaluate the involvement of angiotensinogen(AGT) M235T gene and endothelial nitric oxide synthase(eNOS) G894Tgene polymorphism in the development of essential hypertension(EH).Methods The study included 224 subjects(124 patients with EH and 100 controls).A gene chip technology was used to detect the level of AGT and eNOS gene polymorphisms.Comparison of EH patients vs.the controls in allele frequency and genotype distribution was conducted. Results A significantly higher frequency of the genotype of AGT TT gene was observed in the EH group(74.19% vs.55.00%,χ~2=9.04,P<0.01).The gene polymorphism correlated positively to EH.No statistically significant difference was observed in eNOS GT+TT genotypes distribution between the two groups(12.90% vs.15.00%,P>0.05).Conclusion The AGT-TT gene polymorphism,not eNOS G894T,may be an independent risk factor of EH in China.Gene chip technology provides an effective and sensitive method to study the relationship of gene polymorphism and EH.
