二仙汤抗骨质疏松有效组分对维甲酸致骨丢失大鼠的影响

摘    要::目的 探讨二仙汤抗骨质疏松有效组分对维甲酸致骨质疏松大鼠的影响。方法 60只雌性SD大鼠按体质量随机分为空白对照组、模型组、戊酸雌二醇(0.2mg/kg)组及二仙汤抗骨质疏松有效组分低、中、高剂量(50、100、200mg/kg)组。除空白对照组外,连续14d灌胃给予各组大鼠维甲酸70mg·kg-1·d-1诱导骨质疏松症。造模结束后,按方案对大鼠每天灌胃给药,连续28天。用ELISA试剂盒检测大鼠血清和尿液的生化指标,Micro-CT分析股骨远端骨密度和骨小梁微结构的变化。结果 二仙汤抗骨质疏松有效组分可降低维甲酸骨质疏松大鼠尿液钙的含量(P<0.05),降低血清碱性磷酸酶(P<0.05)和抗酒石酸酸性磷酸酶的活性(P<0.01),增加骨密度,改善骨小梁微结构的退化。结论 二仙汤抗骨质疏松有效组分可减少维甲酸致骨质疏松大鼠的骨丢失和骨组织微结构的退化。

Abstract:Objective To explore effects of antiosteoporotic active fraction from Er-xian decoction (EXD) on osteoporotic rats induced by retinoic acid. Methods Sixty female SD rats were randomly divided as normal control group, model group, estradiol valeratse (0.2mg/kg) treated group and antiosteoporotic active fraction from EXD treated group (50, 100, 200mg/kg). The rats except for animals in normal group were treated with retinoic acid 70mg·kg-1 daily for 14 days by intragastric application to induce osteoporosis. Then, rats were treated with estradiol valeratse and antiosteoporotic active fraction from EXD for consecutive 28 days according to experimental protocol. The biochemical indicators in serum and urine of rats were detected by ELISA kit. The bone density and microstructure of trabecular for distal femur was analyzed with micro-CT. Results: The antiosteoporotic active fraction from EXD decreased calcium content in urine(P<0.05), activities of alkaline phosphatase(P<0.05) and tartratse-resistant acid phosphatase(P<0.01) in serum, significantly increased bone mineral density of femur and improved the microstructure of trabecular in retinoic acid-induced osteoporotic rats. Conclusion Antiosteoporotic active fraction from EXD reduces bone loss and deterioration of bone microarchiateure in retinoic acid-induced osteoporotic rats.
