脑梗死与C反应蛋白基因多态性关系的实验研究

目的研究5个C反应蛋白（CRP）基因rs3122012、rs3116653、rs3116654、rs1130864、rs1205的单核苷酸多态性（SNP）位点与脑梗死的关系。方法采用聚合酶链反应-限制性内切酶片段长度多态性（PCR-RFLP）技术对150例脑梗死患者和125名对照者的CRP基因多态性进行检测,同时检测三酰甘油（TG）、总胆固醇（TC）、高密度脂蛋白胆固醇（HDL-C）、低密度脂蛋白胆固醇（LDL-C）、和血糖（Glu）,并计算非高密度脂蛋白胆固醇（non-HDL-C）。结果脑梗死组患者的收缩压、舒张压、LDL-C和Glu明显高于对照组（P〈0.01）,而TG、TC、HDL-C和non-HDL-C结果2组间差异无统计学意义（P〉0.05）。CRP rs3122012、rs3116653、rs3116654在上海地区人群中无多态性出现,rs1130864有CC、CT和TT型3种基因型,rs1205有AA、AG和GG型3种基因型,但脑梗死组患者的基因型和等位基因频率分布与对照组相比差异无统计学意义（P〉0.05）。结论脑梗死患者可能与高血压、高Glu等因素有关,与CRP基因rs3122012、rs3116653、rs3116654、rs1130864、rs1205的多态性无明显相关。

Research on the correlation between cerebral infarction and C reactive protein gene polymorphism

Objective To investigate the relationship of cerebral infarction with C reactive protein（CRP） gene rs3122012,rs3116653,rs3116654,rs1130864 and rs1205 single nucleotide polymorphism（SNP） point.Methods The polymerase chain reaction-restriction fragment length polymorphism（PCR-RFLP） was used to determine the CRP gene polymorphism in 150 patients with cerebral infarction and 125 controls.The triglyeride（TG）,total cholesterol（TC）,high-density lipoprotein cholesterol（HDL-C）,low-density lipoprotein cholesterol（LDL-C） and glucose（Glu） were determined.The non-HDL-C was calculated.Results The levels of systolic pressure,diastolic pressure,LDL-C and Glu were higher in patients with cerebral infarction than in the controls（P0.01）.There was no statistical difference in TG,TC,HDL-C and non-HDL-C between the 2 groups.There was no polymorphism among CRP rs3122012,rs3116653 and rs3116654 in the population of Shanghai.The rs1130864 had CC,CT and TT genotypes,and the rs1205 had AA,AG and GG genotypes.The genotypes and the allele frequency distribution in cerebral infarction group had no statistical significance compared with those in the control group（P0.05）.Conclusions There is no relationship between CRP gene rs3122012,rs3116653,rs3116654,rs1130864 and rs1205 polymorphism,and the occurrence of cerebral infarction may be related to hypertension and hyperglycemia.
