A－LAK细胞与LAK细胞体外增殖及抗肿瘤作用的比较

本实验观察了Ａ－ＬＡＫ细胞的体外扩增及其抗肿瘤作用，并与ＬＡＫ细胞进行了比较。结果表明，Ａ－ＬＡＫ细胞在短期内大量扩增，在培养第７天时达到增殖高峰，Ａ－ＬＡＫ细胞增加１２．０２倍，而ＬＡＫ细胞增加２．６９倍，培养３周后Ａ－ＬＡＫ细胞增加５３．５０倍，而ＬＡＫ细胞仅增加４．４５倍。１８小时ＬＤＨ－Ｌ释放试验表明，Ａ－ＬＡＫ细胞对Ａｎｉｐ９７３和Ｋ５６２细胞均显示较高的杀伤活性，与ＬＡＫ细胞相比有显著差异（Ｐ＜０．０５）。通过ＦＡＣＳ细胞表型分析显示，Ａ－ＬＡＫ细胞大量表达ＣＤ１６、ＣＤ５６抗原，提示Ａ－ＬＡＫ细胞可能来源于ＬＧＬ－ＮＫ细胞群。

Expansion of A-LAK and LAK Cells in Vitro and Comparison of Its Antitumorigenesis

On this experiment,we have observed the expansion and antitumor activity of A-LAK cells in vitro.The results indicated that A-LAK cells expanded rapidly in the presence of IL-2 in a short culture and the expansion indices reached up to climax on day 7 in the culture.At the same time,A-LAK cells increased 12.02-fold compared with only 2.69-fold for LAK cells.After 3 weeks A-LAK increased 53.5-fold versus 4.45-fold for LAK cells.The results also confirmed that A-LAK had a broad antitumor activity,and that the cytotoxicity of A-LAK against NK-resistant tumor cells(Anip-973) and NK-sensitive tumor cells(K562)was much higher than that of LAK cells by a 18-hour LDH-L release assay,suggesting that there was a significant difference between A-LAK cells and LAK cells(P<0.05)in the cytotoxicity against tumor cells.The phentype analysis of cells by FACS indicated that A-LAK cells express a phenotype characteristic of NK cells with a large number of CD16,CD56 antigen,suggesting that A-LAK cells may stem from LGL-NK population.
