摘    要:本实验观察了A-LAK细胞的体外扩增及其抗肿瘤作用,并与LAK细胞进行了比较。结果表明,A-LAK细胞在短期内大量扩增,在培养第7天时达到增殖高峰,A-LAK细胞增加12.02倍,而LAK细胞增加2.69倍,培养3周后A-LAK细胞增加53.50倍,而LAK细胞仅增加4.45倍。18小时LDH-L释放试验表明,A-LAK细胞对Anip973和K562细胞均显示较高的杀伤活性,与LAK细胞相比有显著差异(P<0.05)。通过FACS细胞表型分析显示,A-LAK细胞大量表达CD16、CD56抗原,提示A-LAK细胞可能来源于LGL-NK细胞群。

Abstract:On this experiment,we have observed the expansion and antitumor activity of A-LAK cells in vitro.The results indicated that A-LAK cells expanded rapidly in the presence of IL-2 in a short culture and the expansion indices reached up to climax on day 7 in the culture.At the same time,A-LAK cells increased 12.02-fold compared with only 2.69-fold for LAK cells.After 3 weeks A-LAK increased 53.5-fold versus 4.45-fold for LAK cells.The results also confirmed that A-LAK had a broad antitumor activity,and that the cytotoxicity of A-LAK against NK-resistant tumor cells(Anip-973) and NK-sensitive tumor cells(K562)was much higher than that of LAK cells by a 18-hour LDH-L release assay,suggesting that there was a significant difference between A-LAK cells and LAK cells(P<0.05)in the cytotoxicity against tumor cells.The phentype analysis of cells by FACS indicated that A-LAK cells express a phenotype characteristic of NK cells with a large number of CD16,CD56 antigen,suggesting that A-LAK cells may stem from LGL-NK population.
