纳米级水镁石纤维粉尘的体外细胞毒性研究

摘    要:目的研究纳米级水镁石纤维粉尘的体外细胞毒性。方法收集大鼠肺巨噬细胞,培养后分别加入纳米级水镁石纤维粉尘、普通水镁石纤维粉尘、石英粉尘及生理盐水,测定肺巨噬细胞存活率、乳酸脱氢酶(LDH)活力。结果纳米级水镁石纤维粉尘的细胞存活率与对照组、普通水镁石纤维粉尘组比较明显降低,差异均有显著性(P<0.05或P<0.01),而与石英组比较明显升高,差异有显著性(P<0.01);LDH实验结果的变化与细胞存活率类似。结论纳米级水镁石纤维粉尘具有明显的细胞毒性,其细胞毒作用较普通水镁石纤维粉尘强,较石英粉尘弱。

Abstract:Objective To study the cytotoxicity of millimicron brucite fiber dust in vitro. Methods The alveolar macrophages of rat lungs were collected , cultured, and then respectively exposed to millimicron brucite fiber, common brucite fiber,quartz dust and 0.9%NaCl respeetively. Their survival rates, activities of lacticdehydrogenase (LDH) were determined. Results Compared with the negative control group and common brucite fiber,the cells survival rate of those exposed to millimicron brucite fiber significantly decreased (P< 0.05 or P< 0.01),but that increased obviously when compared with those of quartz dust(P < 0.01); the changes of the activities of LDH were similar to those of the cells survival rates. Conclusion The cytotoxicity of millimicron brucite fiber was obviously stronger than common brucite fiber, but much weaker than quartz dust.
