| 摘 要： | 目的 观察枸杞多糖对体外培养的雄激素非依赖型人前列腺癌PC-3细胞存活率、细胞周期及其凋亡的影响。方法 用四甲基偶氮噻唑蓝（MTT）法检测枸杞多糖对PC-3细胞作用的时间效应和剂量效应，并计算细胞生长抑制率；流式细胞仪观察枸杞多糖处理PC-3细胞后细胞周期以及细胞凋亡的改变。结果 枸杞多糖对PC-3细胞的增殖有明显的抑制作用，抑制率可达87．84％，且呈剂量-效应和时间-效应关系；流式细胞仪分析显示。枸杞多糖可影响该细胞周期并引起细胞凋亡，凋亡率高达40％。结论 枸杞多糖对人前列腺癌PC-3细胞的生长有明显抑制作用，其机制可能是通过诱导人前列腺癌细胞凋亡和影响其生长周期而实现。

| Abstract: | Objective To investigate the influence of lycium barbarum polysaccharides(LBP)on human prostatic cancer cell line PC-3.Methods Time and dose effects of LBP were examined by using trypan blue exclusion staining;the influence on the cell cycle and the apoptosis were observed with flow cytometry.Single cell gel electrophoresis assay was applied to determine the DNA damage of PC-3 cell.Results LBP could inhibit the growth of PC-3 cell line with a dose-effect and time-effect relationship.LBP could influence the cell cycle of PC-3 and induce apoptosis.The apoptosis rate was 40%.The breakage of DNA strand of PC-3 cell was observed.Conclusion LBP inhibits PC-3 cell line independent of androgen in vitro,and the mechanism may be related to its influence on the cell cycle and inducing of apoptosis. |
