鲍曼不动杆菌基因同源性研究

摘    要:目的分析鲍曼不动杆菌的基因同源性,并对其进行分子分型。方法采用重复序列聚合酶链反应技术(rep—PCR),分别以基因外重复回文序列(REP)和肠杆菌基因间重复一致序列(ERIC)为引物,对122株鲍曼不动杆菌基因组DNA进行扩增并分型。结果REP—PCR将鲍曼不动杆菌分为14种基因型(A~N)。其中H型含98株,为主要的流行型别,并可分为6个亚型;ERIC—PCR将菌株分为13种基因型(A’~M’),其中L’型100株,为主要的流行型别,并可分为5个亚型。其余菌株在其他基因型中均为零星分布。结论在鲍曼不动杆菌基因同源性分析中,REP—PCR的分辨率较ERIC—PCR高;同时表明我院存在着某一基因型鲍曼不动杆菌的感染流行,估计该型菌株可能以克隆株的形式播散。

Abstract:Objective To investigate the gene homology of Acinetobacter baumannii in our hospital and their typing.Methods The genomic DNA of 122 strains were analyzed by repetitive element polymerase chain reaction(rep-PCR) using repetitive extragenic palindromic(REP) and enterobacterial repetitive intergenic consensus sequence(ERICS). Results There were 14 genotypes(A to N) in all isolates with REP-PCR mainly including type H of 98 strains classified to 6 subtypes and 13 genotypes(A' to M') with ERIC-PCR mainly including type L' of 100 strains classified to 5 subtypes,respectively.The distribution of remaining strains were sporadic in the other genotypes. Conclusion The power of REP-PCR is higher than that of ERIC-PCR in discriminating typing of the Acinetobacter baumannii.In addition,in our hospital there was the prevalence of a certain genotype of Acinetobacter baumannii,and it can be clonal dissemination.
