姜黄素对大鼠肺微血管平滑肌细胞周期和增殖能力的影响

摘    要:目的探讨姜黄素对肺微血管平滑肌细胞增殖的影响,及其分子机制。方法原代SD雄性大鼠肺微血管平滑肌细胞的培养、鉴定、传代,MTT法检测细胞增殖,流式细胞术分析细胞周期,RT-PCR检测mRNA的表达,Westernblot检测蛋白表达。结果姜黄素以浓度依赖的方式抑制肺微血管平滑肌细胞增殖,使细胞周期停滞在G0/G1期。姜黄素处理后细胞内p21和p53的mRNA、蛋白水平表达增高、p300的蛋白表达以及p300介导的乙酰化H3和p53蛋白表达降低。结论姜黄素能够抑制肺微血管平滑肌细胞的增殖,使细胞周期停滞在G0/G1期。其作用可能是通过下调p300活性和上调p53相关信号通路来实现的。

Abstract:Objective To explore the effect of curcumin on the proliferation and cell cycle of pulmonary microvascular smooth muscle cells(PMSMCs) and the possible molecular mechanism.Methods Pulmonary microvascular smooth muscle cells were obtained,identified,cultured.The proliferation of the cells was measured by MTT Colorimetric Assay.Cell cycle was analysised by Flow Cytometry.The expression of mRNA and proteins were detected by RT-PCR and Western blot.Results:Curcum ininhibited the PMSMCs proliferation in a concentration dependent manner.Curcumin arrested the progression of cell cycle at G0/G1 phase.Moreover,curcumin upregulated the expression of p53 and p21 and downregulted the protein level of p300 and the acetylation of H3 and p53 mediated by p300.Conclusion:The results indicate that curcumin can inhibit PMSMCs proliferation and arrest the progression of cell cycle at G0/G1 phase via downregulating p300 activity and upregulating p53 related signaling.
