脂肪乳对大鼠局灶性脑缺血-再灌注所致脑损伤的保护作用

摘    要:目的观察20%脂肪乳对大鼠局灶性脑缺血-再灌注损伤后的脑保护作用。方法雄性Sprague-Dawley大鼠23只,随机分为假手术组6只、模型组6只、脂肪乳组11只。线栓法制备右侧大脑中动脉闭塞2h、再灌注24h模型。再灌注即刻,脂肪乳组经尾静脉注射20%脂肪乳0.2mg/kg,模型组注射等体积等渗盐水。再灌注后24h,观察各组神经功能缺失评分、脑梗死体积及腑水肿程度。结果神经功能缺损评分的中位数,模型组为2分,脂肪乳组为1分,两组比较差异有统计学意义(t=81,P〈0.05)。脂肪乳组脑梗死体积分数为(5.4±4.5)%,显著小于模型组的(20.1±2.4)%;脂肪乳组脑水肿程度为(8±5)%,明显低于模型组(28±9)%,差异均有统计学意义(P〈0.01)。结论脂肪乳对急性局灶性脑缺血-再灌注所致的脑损伤具有保护作用。

Abstract:Objective To observe the protective effect of 20% lipid emulsion on focal cerebral ischemia-reperfusion injury in rats. Methods Twenty-three male Sprague-Dawley rats were randomly allo- cated to sham operation (n =6), model(n =6), and lipid emulsion(n = 11 ) groups. The model of right middle cerebral artery occlusion 2 h and reperfusion for 24 h was induced by suture method. 20% lipid emulsion 0. 2 mg/kg was injected into the caudal vein of the rats in the lipid emulsion group immediately af ter reperfusion. The equal amount of normal saline was injected into that of the caudal vein of the rats in the model group. The neurological deficit score, infarct volume, and degree of brain edema in each group were observed at 24 h after reperfusion. Results The median neurological deficit scores were 2 in the model group and 1 in the lipid emulsion group. There were no significant differences between the 2 groups (t = 81 ,P 〈 0.05 ). The score of cerebral infaret volume in the lipid emulsion group was 5.4 ± 4.5% , and it was significantly less than 20. 1 ± 2.4% in the model group. The degree of brain edema of the lipid emulsion group was 8 ±5%, and it was significantly lower than 28 +9% in the model group. There were significant differences ( P 〈 0.01 ). Conclusion Lipid emulsion has a protective effect for acute focal cerebral ischemia-reperfusion-induced brain injury in rats.
