百合中性多糖对5-FU增效减毒作用及其对体外对肿瘤细胞的抑制作用

摘    要:目的观察百合中性多糖(A4糖)对5-氟尿嘧啶(5-fluorouracil,5-FU)抗肿瘤的增效减毒作用及其对体外培养的SGC-7901人胃腺癌细胞增殖的影响。方法建立移植性S180肿瘤小鼠模型,分别观察A4糖单用以及与5-氟尿嘧啶(5-fluorouracil,5-Fu)联合应用时的外周血细胞变化、抑瘤率、脾及胸腺指数的变化;体外培养SGC-7901细胞,MTT法测定A4糖对SCG-7901细胞增殖的影响。结果 A4糖对经5-Fu化疗所引致血中白细胞数量降低的S180荷瘤小鼠有明显的升高白细胞的作用,对化疗所引起脾脏及胸腺指数下降有显著的改善作用;A4糖对体外培养的SGC-7901细胞的生长亦有明显的抑制作用。结论百合中性多糖(A4糖)可通过提高机体免疫功能,从而增强化疗药的抑瘤效果,同时减轻化疗时出现的毒副作用;对体外培养的SGC-7901的生长亦有一定的抑制其增殖的作用。

Abstract:Objective To observe the effects of neutral polysaccharide from Lily on enhancing efficacy and reducing toxicity of 5 - fluorouracil and proliferation of gastric arcinoma cell line SGC - 7901 in vitro. Methods Transplanting S180 mice tumor models were established so as to observe the changes in peripheral blood cells, tumor inhibiting rate and indexes the spleen and thymus when neutral polysaccharide from Lily was used alone and when used in combination with 5 - fluorouracil respectively. SGC - 7901 cells were cultured in vitro. Theproliferation of SCG - 7901 cell for the ation of neutral polysaccharide from Lily was detected by MTT method. Results The Neutral Poly- saccharide from Lily had marked effect in increasing white cell count and had rectifying effect on the drop of spleen and thymus indexes caused by chemotherapy in S180 tumor - hearing mice. It also obviously inhibited the growth of cultured SGC7901 of human gastric adenocarcinoma. Conclusion The Lily - polysaccharide can enhance the antiumor effect of and reduce the adverse reactions of chemotherapy via improving the immunological function. It can inhibite the growth of cultured SGC -7901 of human gastric adenocarcinoma, and promote their apoptosis.
