不同浓度枸杞多糖对大鼠动脉血管平滑肌细胞增殖的影响

摘    要:目的观察大鼠动脉血管平滑肌细胞株A7r5在高浓度葡萄糖作用下的增殖变化以及不同浓度的枸杞多糖(LBP)对该作用的影响。方法将A7r5细胞培养48h后随机分组:对照组(葡萄糖浓度5mM)、高糖组(葡萄糖浓度10、15、20、25、30、40mM)、高糖(25mM)+LBP 60μg.mL-1组、高糖(25mM)+LBP 80μg.mL-1组、高糖(25mM)+LBP 100μg.mL-1组,以MTS孵育A7r5后,经酶联免疫检测仪测定各组的吸光度值。结果①与对照组(1.331±0.097)相比,高糖组吸光度值均明显升高,且以葡萄糖浓度为25mM组(2.485±0.229)增高为甚,与对照组相比差异均有统计学意义(P<0.01)。②高糖(葡萄糖浓度25mM)诱导的A7r5细胞增殖在48~72h达到高峰。③与高糖组相比,中、高浓度LBP组对A7r5细胞增殖有抑制作用,其差异均有统计学意义(P<0.01)。结论高浓度葡萄糖可促进A7r5细胞增殖,中、高浓度LBP可抑制高糖诱导的A7r5增殖。

Abstract:Objective To observe the proliferation of A7r5 cells when high concentration glucose present and the effect of different concentrations of Lycium barbarum polysaccharides(LBP) on the course.Methods Groups: Control(glucose 5mM),high concentration glucose groups(including 10,15,20,25,30 and 40mM),glucose(25mM) + LBP60μg.mL-1,glucose(25mM) + LBP80μg.mL-1,glucose(25mM) + LBP100μg.mL-1.After co-incubating of A7r5 and MTS,OD reading of each group was measured by enzyme immunoassay detecting instrument.Results(1) The OD readings in high glucose groups were all significantly increased,especially in 25mM group(2.485 ± 0.229),compared with controls(1.331 ± 0.097)(either P <0.01).(2) A7r5 proliferation reached its peak when incubated together with glucose(25mM) for 48-72 hours.(3) Compared with 25mM glucose group,the OD readings of LBP 80μg.mL-1 and 100μg.mL-1 groups were significantly decreased(either P < 0.01).Conclusion High concentration glucose accelerates A7r5 proliferating,and LBP suppresses the proliferation of A7r5 induced by high concentration glucose.
