螺旋藻多糖对肝癌细胞BEL7404增殖的影响

摘    要:目的研究螺旋藻多糖对对肝癌细胞增殖的影响,以探讨其抗肝癌机制。方法体外培养肝癌细胞株BEL7404,用MTT法检测螺旋藻多糖作用于BEL740424h后的细胞增殖情况;流式细胞术(FCM)检测对细胞周期的影响和凋亡率。结果MTT法测定结果表明,螺旋藻多糖对BEL7404增殖有抑制作用;FCM检测表明,螺旋藻多糖引起增殖中的BEL7404细胞发生G1期阻滞。结论螺旋藻多糖能有效地抑制BEL7404细胞增殖,提示阻滞细胞周期、抑制细胞增殖可能是螺旋藻多糖治疗肝癌的一个机制。

Abstract:Objective To observe the effect of polysaccharide from Spirulina platensis on proliferation of BEL7404 cells and to explore its anti-hepatocellular carcinoma mechanisms.Methods Effect of polysaccharide from Spirulina on proliferation of the BEL7404 cell was detected by MTT assay and the cell cycle distribution was detected by flow cytometry.Apoptosis of BEL7404 cell was investigated further.Results Polysaccharide from Spirulina inhibited the proliferation of BEL7404 cell.Polysaccharide from Spirulina could also induce apoptosis,cause the G1 phase block of BEL7404 cell,and the ratio of apoptosis was significantly difference with that of control group.Conclusion Polysaccharide from Spirulina inhibits proliferation of BEL7404 cells via induction of apoptosis and cell cycle arrest.
