高压氧对缺氧缺血新生大鼠脑组织存活素及细胞凋亡蛋白酶-3表达的影响

摘    要:目的 观察高压氧(HBO)对新生大鼠缺氧缺血性脑损伤(hypoxia-ischemia brain damage,HIBD)后脑组织存活素(survivin)及细胞凋亡蛋白酶-3(caspase-3)表达的影响,并探讨其保护机制.方法 144只7 d龄SD大鼠随机分为对照组(n=16)、新生HIBD组(n=64)及HBO治疗组(n=64).采用经典的Rice法制作缺氧缺血性脑损伤模型,应用Epics XL流式细胞仪检测细胞凋亡情况,并应用免疫组织化学法检测Survivin和Caspase-3表达变化.结果 与对照组相比,HIBD组细胞凋亡率及Survivin、Caspase-3表达水平明显升高,组间比较差异有统计学意义(P<0.05).通过相关性分析,HBO治疗可促进存活素表达,其表达水平和caspase-3表达负相关.结论 HBO可能通过增加抗凋亡因子Survivin表达来抑制Caspase-3激活,减少缺氧缺血性脑损伤后细胞凋亡,发挥神经保护作用.

Abstract:Objective To explore the effect of hyperbaric oxygen (HBO) on the expression of survivin and caspase-3 following hypoxia-ischemia brain damage (HIBD) in neonatal rats,and the mechanism involved in the protection of the neural system by HBO.Methods One hundred and forty-four ( aged seven days) rats were randomly divided into three groups,i.e.the control group (n = 16),the HIBD group (n =64) and the HBO group (n =64).An experimental HIBD model was developed by the Rice method.Following damage,samples were obtained for measuring apoptosis of cells with the Epics XL flow cytometer and survivin expression and caspase-3 proteins were measured with immuno-histochemistry.Results When compared with that of the control group,the number of apoptotic cells,survivin,and caspase-3 immunopositive cells all increased significantly in the HBID group,and statistical differences could be seen between groups ( P < 0.05 ).Analyses showed that HBO could significantly enhance the expression of survivin,with negative correlation between the level of survivin expression and the activity of caspase-3.Conclusions Increased expression of survivin by HBO might be one of the mechanisms involved in the regulation of caspase-3 and inhibition of neural cell apoptosis in cortex,which might be protective to neurons.
