变性梯度凝胶电泳和单链构象多态性分析检测皮肤癌p53基因突变的比较

摘    要:目的:检测皮肤癌p53基因5-8外显子的突变频率,比较变性和梯度凝胶电泳(DGGE)和单链构象多态性分析(SSCP)检测p53基因突变的敏感性,方法:分别采用DGGE和SSCP,对40例皮肤癌患者手术切除组织的p53基因5-8外显子进行突变检测。结果:皮肤癌p53基因5-8外显子的总突变率为35%,其中鳞状细胞癌(SCC)的突变率为36%,基底细胞癌(BCC)为33%,采用GGE和SSCP检测的突变率分别为33%和25%。结论35%的皮肤癌组织存在p53基因5-8外显子突变。GGE检测p53基因突变的敏感性高于SSCP。DGGE结合SSCP有助于提高突变检出率。

A Comparison of Denaturing Gradient Gel Electrophoresis and Single Strand Conformation Polymorphism Analysis in Detecting the Mutation of p53 Gene in Skin Cancer
Abstract:Purpose To detect the mutation in the exon 5-8 of p53 gene in skin cancer by denaturing gradient gel electrophoresis (DGGE) and single strand conformation polymorphism (SSCP).Comparing the sensitivity of DGGE and SSCP in screening mutations of p53 gene. Methods The mutation in the exon 5-8 of p53 gene was detected by DGGE and SSCP in 40 patients with skin cancers. Results We found the aberrant band migrations in 13 patients (33%) using DGGE and in 10 patients (25%) using SSCP.The total frequency of mutations in exon 5-8 of p53 gene was 35%.36%(8/22) of the SCC tested presented a mutated p53 gene,the frequency was lower in our series of BCC (33%,6/18). Conclusions The total frequency of mutations in exon 5-8 of p53 gene in skin cancers was 35%.DGGE is more sensitive than SSCP in screening mutations of p53 gene.The combination of DGGE with SSCP can improve the detective rate of mutation.
