Abstract: | IgM subunits were obtained by reduction of the native IgM with different reagents. Upon removal of the reducing agent they reaggregated spontaneously into a variety of molecular species sedimenting from 8 to 17 S. Whereas the choice of the reducting agent did not influence the rate of aggregation, an increase of pH from 7 to 10 and the presence of imidazole tended to inhibit the formation of aggregates. Rate constants for the formation of single aggregates ranged from 3 × 10−6 sec−1 to 2 × 10−5 sec −1 which differs by a factor of 103 from the rate constants found in the subunit reduction of IgM. A mechanism of stepwise formation of polymers of the general formula (μ2κ2)n is discussed. |
